摘 要:目的对山海螺的显微特征、薄层色谱鉴别、水分、总灰分、浸出物和紫丁香苷含量进行试验,为制订山海螺的质量标准提供参考。方法通过显微镜观察山海螺的显微特征;采用薄层色谱方法对山海螺进行定性鉴别;采用高效液相法测定紫丁香苷的含量;测定5批山海螺的水分、总灰分、浸出物和紫丁香苷含量,制定其合理限度。结果山海螺的显微特征明显易见,专属性强;薄层色谱鉴别中,山海螺与对照药材色谱相应的位置上显示的斑点对应整齐;紫丁香苷在0.02248~0.2248μg范围内,与峰面积呈良好的线性关系,加样回收率在96.95%~103.76%范围内。拟定水分和总灰分限度分别为不得超过13.0%和4.0%;以水为溶剂,按热测定法测定,浸出物不得少于53.0%;紫丁香苷含量不得少于0.055mg/g。结论该方法稳定、可靠,可应用于控制山海螺的质量。Objective To conduct experiments on the microscopic characteristics,thin-layer chromatography identification,moisture content,total ash,extractives and syringin content ofCodonopsislanceolata,in order to provide reference for the formulation of quality standards for Codonopsislanceolata.Methods The microscope was used to observe the microscopic characteristics of Codonopsislanceolata.Thin-layer chromatography was used for the qualitative identification of Codonopsislanceolata.High-performance liquid chromatography was used to determine the content of syringin.Moisture content,total ash,extractives,and syringin content in 5 batches of Codonopsislanceolata were determined to establish the reasonable limits.Results The microscopic characteristics of Codonopsislanceolata were clearly visible and have strong specificity.Thin-layer chromatography showed corresponding spots at the same positions for Codonopsislanceolata and the control herb.Syringin demonstrated a good linear correlations with the respective chromatographic peak areas within the ranges of 0.02248-0.2248μg,and the sample recovery rate were within the range of 96.95%to 103.76%.The moisture and total ash were limited to not exceed 13.0%and 4.0%,respectively.Water as the solvent and determined by hot water extraction,the extractives were limited to not be less than 53.0%.Syringin content was limited to not be less than 0.055 mg/g.Conclusion This method is stable,reliable,and can be applied to control the quality ofcodonopsis lanceolata.
