摘 要:目的 探讨补肾调经汤对多囊卵巢综合征(PCOS)模型大鼠中肿瘤坏死因子-α(TNF-α)表达的影响。方法 选取60只SD大鼠作为研究对象,将其随机分为对照组,模型组,中药低、中、高剂量组,西药组,每组各10只。观察各组大鼠体质量变化;采用HE染色观察各组大鼠卵巢组织病理学变化并计算窦卵泡数、颗粒细胞层数及卵泡基底膜厚度;采用ELISA观察各组大鼠血清睾酮(T)、卵泡刺激素(FSH)、促黄体生成素(LH)、雌二醇(E2)水平;采用Western Blot法观察各组大鼠组织中TNF-α表达。结果 大鼠造模前体质量比较差异无统计学意义(P>0.05);造模后,与对照组相比,模型组和中药低、中剂量组大鼠体质量有所升高;与模型组相比,中药低、中、高剂量组和西药组体质量有所降低(P<0.05)。与对照组相比,模型组大鼠窦卵泡数、卵泡基底膜厚度较高,颗粒细胞层数较低;与模型组比较,中药低、中、高剂量组和西药组窦卵泡数、卵泡基底膜厚度降低,颗粒细胞层数升高(P<0.05)。与对照组相比,模型组大鼠血清T、FSH、LH水平较高,E2水平较低;与模型组相比,中药低、中、高剂量组和西药组T、FSH、LH水平有所降低,E2水平有所上升(P<0.05)。与对照组相比,模型组大鼠TNF-α蛋白表达较高;与模型组相比,中药低、中、高剂量组和西药组大鼠TNF-α蛋白表达有所降低(P<0.05)。结论 补肾调经汤可能通过对卵泡膜和颗粒细胞的作用进一步降低PCOS大鼠的雄激素水平,可以有效改善内分泌紊乱,调控TNF-α,促进卵泡成熟并排卵。Objective To investigate the effect of bushen tiaojing decoction on the expression of tumor necrosis factor-α(TNF-α)in rats with polycystic ovary syndrome(PCOS).Methods Sixty SD rats were selected as study subjects and randomly divided into control group,model group,low,medium and high dose group of traditional Chinese medicine,and western medicine group,with 10 rats in each group.The body mass of rats in each group was observed;HE staining was used to observe the histopathological changes in the ovaries of rats in each group and to calculate the number of sinusoidal follicles,the number of granulosa cell layers and the thickness of the follicular basement membrane;ELISA was used to observe the levels of serum testosterone(T),follicle-stimulating hormone(FSH),luteinizing hormone(LH),and estradiol(E2)in the rats in each group;and Western Blot was used to observe TNF-αin the tissues of the rats in each group.Results Comparison of body mass of rats before modeling was not statistically significant(P>0.05);after modeling,compared with the control group,body mass of rats in the model and low and medium dose groups of traditional Chinese medicine was elevated,and body mass of rats in the low,medium and high dose groups of traditional Chinese medicine and the western medicine group was reduced compared with the model group(P<0.05).Compared with the control group,rats in the model group had higher sinus follicle number,follicular basement membrane thickness,and lower granulosa cell layer number;compared with the model group,sinus follicle number,follicular basement membrane thickness decreased,and granulosa cell layer number increased in the low,medium,and high dosages of the traditional Chinese medicine group and the western medicine group(P<0.05).Compared with the control group,the serum levels of T,FSH,and LH were higher and the level of E2 was lower in the model group;compared with the model group,the levels of T,FSH,and LH were lower and the level of E2 was higher in the low,medium and high dose groups of traditi
