摘 要:为了研究菲(Phenanthrene,PHE)对泥鳅(Misgurnus anguillicaudatus)性腺热休克蛋白(Heat shock proteins,HSP)、雌激素受体(Estrogen receptor,ER)及其相关通路的影响,探讨其对生殖内分泌干扰机制,本研究设置4个菲浓度组,采用静水实验和实时荧光定量法分别在4个不同胁迫时间检测泥鳅精巢组织、卵巢组织中HSP基因(包括hsp90α、hsp90β)和ER基因(包括erα、erβ)的mRNA表达量。结果显示,在菲胁迫下,泥鳅精巢组织中hsp90α表达量呈现显著上调,而hsp90β表达量呈现显著下调,两种er的表达均显著上调,其中erβ上调更加明显;在菲胁迫下,泥鳅卵巢组织中hsp90β表达量呈现显著上调,而hsp90α的表达量呈现显著下调,两种er的表达量均显著下调,其中erα下调幅度更大。上述结果说明,菲可能主要通过诱导hsp90α和erβ表达来干扰精巢HSP90-ER-E通路,而通过诱导hsp90β和抑制erα来干扰卵巢HSP90-ER-E通路,从而分别影响雌激素对雄性和雌性泥鳅生殖内分泌调控作用。To investigate the effects of phenanthrene(PHE)on HSP,ER and related pathways in the gonads of Misgurnus anguillicaudatus,and to explore its mechanism of reproductive endocrine disruption,four concentration groups of phenanthrene was established.Using static water experiments and real-time fluorescent quantitative methods,the mRNA expression levels of HSP genes(including hsp90αand hsp90β)and ER genes(including erαand erβ)were measured in the testis and ovary tissues of Misgurnus anguillicaudatus at four different stress durations.The results showed that under PHE stress,the expression of hsp90αwas significantly up-regulated in the testis of Misgurnus anguillicaudatus,while hsp90βwas significantly down-regulated,and the expression of both er genes was significantly up-regulated,among which the up-regulation of erβwas more pronounced;under PHE stress,the expression of hsp90βwas significantly up-regulated in the ovary of Misgurnus anguil⁃licaudatus,while hsp90αwas significantly down-regulated,and the expressions of erαand erβwere significantly down-regulated,among which the down-regulation of erαwas more significant.The above results suggest that PHE may interfere with the spermatophore HSP90-ER-E pathway mainly by inducing the expression of hsp90αand erβ,whereas it may interfere with the ovarian HSP90-ER-E pathway by inducing hsp90βand inhibiting erα,thus affect⁃ing the endocrine reproductive regulation of estrogen in male and female Misgurnus anguillicaudatus.
