清心益智助眠药枕基础方对失眠小鼠心脏、肝脏及脑组织的影响

摘　　要：[目的]本文旨在阐明清心益智助眠药枕基础方(交泰丸、二至丸、聪明汤)对失眠小鼠的药效作用。[方法]以对氯苯丙氨酸(DL-4-chlorophenylalanine, PCPA)为诱导剂,建立小鼠失眠模型,试验期间观察小鼠行为学变化,通过观察小鼠肝脏、心脏、脑组织的病理变化,测定肝脏组织的丙二醛(MDA)含量及超氧化物歧化酶(SOD)活性、心脏组织的去甲肾上腺素(NE)含量、脑组织的乙酰胆碱(Ach)含量等病理生化指标,初步评价药枕基础方对PCPA致失眠模型小鼠的影响。[结果]药枕基础方可改善PCPA致失眠小鼠的肝脏、心脏和脑组织病理变化,模型组小鼠肝脏组织SOD活性、脑组织Ach含量较空白组显著降低(P<0.05),模型组小鼠肝组织MDA含量、心组织NE含量较空白组显著升高,说明造模成功。药枕基础方组与阳性对照组小鼠肝组织SOD活性和脑组织Ach含量均显著高于模型组;药枕基础方组与阳性对照组小鼠肝组织MDA含量和心组织NE含量均显著低于模型组。[结论]药枕基础方可有效改善PCPA致失眠小鼠的睡眠质量,显著提高肝组织SOD活性、脑组织的Ach含量,降低肝组织MDA含量以及心组织的NE含量。[Objectives]The paper was to elucidate the efficacy of the basic formula of Qingxin nootropic sleep aid pillow(Jiaotai pill,Erzhi pill and smart decoction)on insomnia mice.[Methods]The DL-4-chlorophenylalanine(PCPA)was used as an inducer to establish a mouse insomnia model,and the behavioral changes of mice were observed during the experiment,and the pathological and biochemical indexes such as malondialdehyde(MDA)content and superoxide dismutase(SOD)activity in liver tissue,norepinephrine(NE)content in heart tissue and acetylcholine(Ach)content in brain tissue were measured by observing the pathological changes of heart,liver and brain tissues.The effect of the basic formula of drug pillow on PCPA-induced insomnia model mice was preliminarily evaluated.[Results]The basic prescription of the drug pillow could improve the pathological changes of heart,liver and brain tissues of PCPA-induced insomnia mice.The SOD activity and Ach content in liver tissue of mice in the model group were significantly lower than those in the blank group(P<0.05),and the MDA content in the liver tissue and NE content in the heart tissue of the model group were significantly higher than those in the blank group,indicating that the modeling was successful.The SOD activity in liver tissue and Ach content in brain tissue of mice in the basic formula group and the positive control group were significantly higher than those in the model group.The MDA content in liver tissue and NE content in heart tissue of mice in the basic formula group and the positive control group were significantly lower than those in the model group.[Conclusions]The basic prescription of the drug pillow could effectively improve the sleep quality of PCPA-induced insomnia mice,and significantly increase the SOD activity in liver tissue,the content of Ach in brain tissue,and reduce MDA content in liver tissue and the content of NE in heart tissue.
