摘 要:为探讨甜菜碱(BET)对热应激山羊胎盘滋养层细胞(GTCs)增殖凋亡及抗氧化功能的影响,试验利用不同浓度甜菜碱处理体外培养的GTCs,采用CCK-8法检测细胞活力、采用流式细胞术检测细胞凋亡率、采用比色法检测细胞总抗氧化能力(T-AOC)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)活性及丙二醛(MDA)含量等细胞抗氧化指标。结果发现:在正常培养条件下,添加1 mmol/L甜菜碱能够显著增强GTCs的细胞活力(P<0.05),而25 mmol/L甜菜碱使细胞活力极显著降低(P<0.01)。GTCs在42℃处理12 h后,热应激组(HS)GTCs的细胞活力极显著降低;添加1 mmol/L甜菜碱能够显著增强细胞在热应激培养条件下的活力(P<0.01)、显著降低热应激诱导的细胞凋亡(P<0.01);添加1 mmol/L甜菜碱显著降低了热应激培养条件下GTCs中脂质过氧化反应产物丙二醛的含量(P<0.05)。由此可知,甜菜碱可通过抑制热应激条件下GTCs中的过氧化反应而缓解热应激诱导的细胞活力下降和异常细胞凋亡。In order to investigate the effects of betaine(BET)on the proliferation,apoptosis and antioxidant function of goat trophoblast cells(GTCs)under heat stress,the GTCs were treated with different concentrations of betaine.Cell viability was detected by Cell Counting Kit-8(CCK-8),cell apoptosis rate was detected by flow cytometry,total antioxidant capacity(T-AOC),the activities of superoxide dismutase(SOD),catalase(CAT)and malondialdehyde(MDA)were detected by colorimetry.The results showed that the addition of 1 mmol/L betaine significantly increased the cell viability of GTCs(P<0.05),while 25 mmol/L betaine sig-nificantly decreased the cell viability(P<0.01).After GTCs were treated at 42℃for 12 h,the cell viability of GTCs decreased significantly in heat stress(HS)group.However,the addition of 1 mmol/L betaine could sig-nificantly enhance the viability of cells under heat stress(P<0.01),and significantly reduce the apoptosis induced by heat stress(P<0.01).1 mmol/L betaine significantly decreased the content of malondialdehyde(MDA),a key product of lipid peroxidation,in GTCs under heat stress(P<0.05).It was concluded that betaine the peroxidation reaction in GTCs under heat stress conditions.
