摘 要:试验旨在分析金银花叶与连翘叶提取物作为饲料添加剂中抗生素替代物的应用潜力。采用2,4,6-三硝基苯磺酸(TNBS)诱导炎症性肠病斑马鱼,分别考察金银花叶水提物(WEHL组)、连翘叶水提物(WEFL组)、金银花叶与连翘叶水提物(WEHL+WEFL组)及土霉素(OXY组)对斑马鱼肠组织形态、中性粒细胞表达及炎性细胞因子水平的影响,并评价其抗炎效果。结果显示:与Control组相比,TNBS组斑马鱼肠道长度显著缩短(P<0.05),宽度显著增加(P<0.05);与TNBS组相比,各给药组斑马鱼肠道长度显著增长(P<0.05),宽度显著降低(P<0.05),WEHL+WEFL组肠道扩张效果最好。与Control组相比,TNBS组肠道中性粒细胞显著增多(P<0.05)。与TNBS组相比,各给药组中性粒细胞数量显著减少(P<0.05)。与TNBS组相比,各给药组斑马鱼中性粒细胞及炎性细胞因子肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)的表达量均显著降低(P<0.05),WEHL+WEFL组与OXY组结果更接近,表现出良好的抗炎效果。综上所述,金银花叶与连翘叶水提物能显著改善炎性肠病斑马鱼的肠道损伤,降低炎性细胞因子的表达,抗炎效果显著,在饲料抗生素替代方面有较好的应用潜力。The experiment aims to analyze the potential application of extracts from honeysuckle leaves and forsythia leaves as alternatives to antibiotics in feed additives.Using 2,4,6-trinitrobenzenesulfonic acid(TNBS)to induce inflammatory bowel disease in zebrafish,the effects of honeysuckle leaf extract(WEHL),forsythia leaf extract(WEFL),honey-suckle leaf and forsythia leaf extract(WEHL+WEFL),and positive drug oxytetracycline(OXY)on the intestinal tissue morphology,intestinal structure and inflammatory cytokine levels of zebrafish were investigated,and their anti-inflammatory effects were evaluated.The results showed that compared with the control group,the TNBS group significantly shortened the intestinal length of zebrafish(P<0.05)and significantly increased the width(P<0.05).Compared with the TNBS group,the zebrafish intestinal length significantly increased(P<0.05)and the width significantly decreased(P<0.05)in each treatment group,with the WEHL+WEFL group showing the best intestinal dilation effect.Compared with the control group,the TNBS group showed a significant increase in intestinal neutrophils(P<0.05).Compared with the TNBS group,the number of neutrophils in each treatment group was significantly reduced(P<0.05).Compared with the TNBS group,the expression levels of neutrophils and inflammatory cytokines TNF-α,IL-1β,IL-6 in zebrafish in each treatment group were significantly reduced(P<0.05).The results of the WEHL+WEFL group were closer to those of the positive drug OXY group,showing good anti-inflammatory effects.Water extracts of honeysuckle leaves and Forsythia suspensa leaves can signifi-cantly improve intestinal damage in zebrafish with inflammatory bowel disease,reduce the expression of inflammatory cytokines,and have significant anti-inflammatory effects.They have good potential for applica-tion in the replacement of feed antibiotics.
