摘  要:为建立无(少)侧芽切花菊组培快繁技术体系,筛选对其芽诱导、增殖和生根的最佳培养基配方,本试验以切花菊茎段为试验材料,使用75%乙醇和0.5%苯扎溴铵进行消毒,筛选出适宜菊花茎段的消毒方法;设置不同激素配比,对茎段接种诱导率、褐化率等,不定芽增殖系数以及组培苗生根率进行研究。结果表明,75%酒精消毒20 s+0.5%苯扎溴铵15 min消毒效果最佳,存活率高达96.0%;A5培养基(MS+6-BA 1.0 mg/L+NAA 0.5 mg/L+蔗糖30 g/L+琼脂6 g/L)是芽诱导最佳培养基,诱导率为78.33%;B1培养基(MS+6-BA 0.5 mg/L+NAA 0.2 mg/L+蔗糖30 g/L+琼脂6 g/L)是增殖最佳培养基,增殖系数为4.88;C3培养基(1/2 MS+IBA 0.1 mg/L+NAA 0.1 mg/L+蔗糖20 g/L+琼脂6 g/L)是生根最佳培养基,生根率为100%。本文为无(少)侧芽切花菊工厂化生产提供参考。To establish the tissue culture and rapid propagation technology system of cut flower chrysanthemum group without(few)side buds,and the best medium formula for bud induction,proliferation and rooting was screened,the stem segment of cut flower chrysanthemum was used as the test material,and the stem segment was disinfected with 75%alcohol and 0.5%benzalkonium bromide,and the appropriate disinfection method was selected.The induction rate,browning rate,coefficient of adventitious bud proliferation,and rooting rate of tissue culture seedlings were studied by setting different hormone ratios.The results showed that 75%alcohol disinfection for 20 s+0.5%benzal konium bromide for 15 min had the best disinfection effect,and the survival rate was as high as 96.0%.A5 medium(MS+6-BA 1.0 mg/L+NAA 0.5 mg/L+sucrose 30 g/L+agar 6 g/L)was the best medium for bud induction,and the induction rate was 78.33%.B1 medium(MS+6-BA 0.5 mg/L+NAA 0.2 mg/L+sucrose 30 g/L+agar 6 g/L)was the best medium for proliferation,and the proliferation coefficient was 4.88.C3 medium(1/2 MS+IBA 0.1 mg/L+NAA 0.1 mg/L+sucrose 20 g/L+agar 6 g/L)was the best medium for rooting,and the rooting rate was 100%.This paper provides references for the factory production of chrysanthemum without(less)side bud cutting.
