基于生物信息学与网络药理学探析祛湿解毒颗粒治疗艾滋病免疫重建不全的作用机制

摘　　要：目的 本文旨在通过生物信息学与网络药理学相关分析,探析祛湿解毒颗粒治疗艾滋病免疫重建不全的作用靶点以及作用机制。方法 使用GEO数据集数据库中的GSE77939、GSE106792数据集以及Gene Cards数据库收集艾滋病免疫重建不全相关疾病靶点;运用ETCM V2.0数据库获取祛湿解毒颗粒活性成分及靶点;STRING数据库分析蛋白相互作用;R软件与Cytoscape软件进行数据分析及可视化,并进行GO分析与KEGG分析。结果 共得到疾病靶点1 392个,药物靶点333个,“疾病-药物”交集靶点97个,KEGG富集分析共富集到通路141条,其中与艾滋病、免疫相关通路8条。结论 祛湿解毒颗粒可通过增加CD4细胞增殖、分化与减少CD4细胞凋亡两方面促进艾滋病免疫重建不全患者免疫重建。Objective This study aimed to explore the target and mechanism of Qushi Jiedu Granules for treating immunological non-responders of AIDS through bioinformatics and network pharmacology.Methods The GSE77939 and GSE106792 datasets in the Gene Expression Omnibus and Gene Cards databases were used to identify targets of AIDS-related diseases among immunological non-responders.The active components and targets of Qushi Jiedu granules were obtained from the ETCM V2.0 database.The STRING database was used to analyze protein interactions.The R and Cytoscape software were used for data analysis and visualization,and Gene Ontology and Kyoto Encyclopedia of Genes and Genomes(KEGG)analyses were performed.Results A total of 1392 disease targets,333 drug targets,and 97 disease–drug intersection targets were obtained.A total of 141 pathways were enriched in the KEGG enrichment analysis including eight pathways related to AIDS and immunity.Conclusions Qushi Jiedu granules could promote immune reconstruction in immunological non-responders of AIDS by increasing the proliferation and differentiation of CD4 cells and reducing the apoptosis of CD4 cells.
