和厚朴酚对水霉菌丝的体外抑制效果研究

摘  要:目的水霉病是一种由丝状真菌所引起的水产动物常见真菌性疾病,通过感染水产动物体表受伤组织及鱼卵,导致水产动物出现高死亡率和低孵化率,给我国乃至全世界水产养殖业带来巨大的经济损失.研究和厚朴酚的不同质量浓度的提取液对水霉菌丝的体外抑制效果,并通过菌丝超微结构变化情况、观测、分析其潜在的作用机制.方法试验应用二倍稀释的方法依次配成512 mg/L、256 mg/L、128 mg/L、64 mg/L、32 mg/L、16 mg/L、8 mg/L、4 mg/L、2 mg/L、1 mg/L的含药物培养基,经高温杀菌后在十二孔板中依次进行已培养好的水霉菌丝,待药物作用24 h后,观察水霉菌丝萌发情况,找出对菌丝抑制作用最佳的质量浓度.结果电镜观察显示32 mg/L和厚朴酚处理的水霉菌丝细胞损伤相对严重,呈现重度水肿现象;细胞内基质稀疏、变淡,细胞器肿胀;细胞膜局部破损、凹陷并质壁分离.说明适宜质量浓度的和厚朴酚对水霉菌丝胞内结构造成了明显损伤,从而对其生长萌发具有显著的抑制效果.结论当厚朴酚质量浓度大于32 mg/L时,药物对水霉菌丝的抑制杀灭有明显的效果,和厚朴酚具有成为水霉病防治药物的潜力,为和厚朴酚作为水霉病防治药物提供科学依据.Objective Saprolegniasis is a common fungal disease of aquatic animals caused by filamentous fungi,which leads to high mortality and low hatching rate of aquatic animals by infecting injured tissues on the body surface of aquatic animals and fish eggs,and brings great economic losses to the aquaculture industry in our country and even all over the world.In this paper,the inhibitory effect of different concentrations of honokiol on water mycelial filaments in vitro was investigated,and the potential mechanism of action was analyzed by observing the changes of mycelial ultrastructure.Methods Experiments were carried out by applying two-fold dilution method to formulate 512 mg/L,256 mg/L,128 mg/L,64 mg/L,32 mg/L,16 mg/L,8 mg/L,4 mg/L,2 mg/L,1 mg/L medium containing the drug in order,and then the cultured Saprolegnia was re-strengthened in twelve-well plates after high temperature sterilisation,and then the drug was observed after 24 h.The germination situation of Saprolegnia filaments was found out.After 24 h of drug action,we observed the hypha germination situation to find out the best concentration for mycelial inhibition.Results Electron microscope observation showed that the cell damage of Saprolegnia treated with 32 mg/L honokiol was relatively serious,presenting the phenomenon of heavy oedema;the intracellular matrix was sparse and pale and the organelles were swollen;the cell membranes were locally broken,concave and the plasma wall was separated.It indicated that the appropriate concentration of Saprolegnia and honokiol caused obvious damage to the intracellular structure of Saprolegnia filaments and thus had a significant inhibitory effect on their growth and germination.Conclusion when the concentration of honokiol is greater than 32 mg/L,the drug has obvions inhibition and killing effect on Saprolegnia hypha Honokiol has the potential to be used as a drug for Saprolegnia control and this study provides a scientific basis for honokiol as a drug for treating saprolegniasis.
