摘 要:目的通过生物信息学方法识别多囊卵巢综合征(PCOS)和干燥综合征(SS)共同的差异表达基因(DEGs),并探讨相关的分子机制。方法从GEO数据库中下载3个PCOS数据集(GSE156067、GSE12422、GSE5090)和3个原发性干燥综合征(pSS)数据集(GSE66795、GSE84844、GSE51092),获取PCOS/pSS与正常对照组的基因表达数据。用limma包进行分析,获得PCOS与pSS之间的DEGs,进而进行功能富集分析和信号通路分析,构建蛋白-蛋白相互作用网络(PPI),使用STRING和Cytoscape软件鉴定枢纽(Hub)基因。结果从上述数据集中共筛选出20个DEGs,GO和KEGG富集分析表明DEGs主要与免疫、炎症和感染相关;进而从中筛选出与PCOS/pSS相关的8个Hub基因(包括CCL2、EGR1、PTGS2、IL15、MAFB、CD83、OASL、ISG20)。结论多囊卵巢综合征与干燥综合征可能存在相关的发病途径,本研究为两疾病的诊治提供了新思路和可能的生物标志物。Objective The present study aims to identify the differentially expressed genes(DEGs)between PCOS and Sjögren syndrome(SS)through the bioinformatics method,and explore the associated molecular mechanisms.Methods We obtained gene expression data of PCOS/SS patients and normal controls from three PCOS databases(GSE156067,GSE12422,GSE5090)and three primary Sjögren syndrome(pSS)databases(GSE66795,GSE84844,GSE51092).The diferentially expressed genes(DEGs)between PCOS and pSS were obtained by the limma R package analysis,followed by functional enrichment analysis and signaling pathway analysis,protein-protein interaction network(PPI)constraction,and Hub genes were identified using STRING and Cytoscape software.Results A total of 20 DEGs were extracted from the above dataset.GO and KEGG enrichment analysis showed that DEGs was mainly enriched in immune,inflammatory and infection.And then,8 Hub genes(CCL2,EGR1,PTGS2,IL15,MAFB,CD83,OASL and ISG20)were selected,which was related with PCOS/pSS.Conclusion Polycystic ovary syndrome and Sjögren syndrome may have relevant pathogenesis,which provides novel insights and possible biomarkers for the diagnosis and treatment of the two diseases.
