石斛多糖对小鼠体内血糖、血脂浓度、免疫功能及抗疲劳能力的影响

摘  要:为探究石斛多糖(Dendrobium polysaccharides,DP)对Ⅱ型糖尿病小鼠降低血糖和血脂浓度、免疫低下小鼠免疫调节及正常小鼠抗疲劳能力的影响,将小鼠平均且随机分成6组,分别命名为空白组、模型组、阳性组、石斛多糖高剂量组(DP-H组,450 mg/kg)、石斛多糖中剂量组(DP-M组,300 mg/kg)、石斛多糖低剂量组(DP-L组,150 mg/kg)。Ⅱ型糖尿病小鼠模型的构建采用腹腔注射四氧嘧啶(alloxan monohydrate,ALX,100 mg/kg)的方法,阳性组使用盐酸二甲双胍(metformin hydrochloride,MET,200 mg/kg),各组灌胃3周后,测定小鼠空腹血糖浓度、血清中总胆固醇(to-tal cholesterol,TC)、甘油三酯(triglyceride,TG)、低密度脂蛋白胆固醇(low-density lipoprotein cholesterol,LDL-C)、高密度脂蛋白胆固醇(high-density lipoprotein cholesterol,HDL-C)含量;免疫低下小鼠模型的构建采用腹腔注射环磷酰胺(cyclophosphamide,CTX,80 mg/kg)的方法,分组同上,阳性组使用左旋咪唑(40 mg/kg),各组灌胃3周后,测定小鼠免疫器官指数及细胞免疫因子含量;正常小鼠分组同上,阳性组使用牛磺酸(200 mg/kg),各组灌胃4周后,记录小鼠负重游泳时间,测定尿素氮(blood urea nitrogen,BUN)、乳酸(lactic acid,LD)含量及肌酸激酶(creatine kinase,CK)、超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)活性。结果表明,相比于模型组,经DP灌胃后,小鼠空腹血糖浓度、TC含量、TG含量、LDL-C含量均有不同程度的降低,而HDL-C含量升高;免疫调节实验中,DP组胸腺及脾脏指数、细胞免疫因子含量均呈现升高的趋势,且呈现剂量依赖性;DP-H组负重游泳时间、GSH-Px活性、SOD活性显著提高(P<0.05),CK活性、BUN含量、LD含量降低。综上所述,石斛多糖可平衡血糖浓度,促进血脂代谢,增强免疫功能,并具有提高抗疲劳能力的作用。In order to explore the effects of Dendrobium polysaccharides(DP)on the reduction of blood glucose and blood lipid concentration in mice with type II diabetes mellitus,immunoregulation in immunocompromised mice,and anti-fatigue ability of normal mice,the mice were randomly divided into six groups,which were blank group,model group,positive group,high-dose DP group(DP-H,450 mg/kg),mediumdose DP group(DP-M,300 mg/kg),and low-dose DP group(DP-L,150 mg/kg).Intraperitoneal injection of alloxan monohydrate(ALX,100 mg/kg)was used to construct a mouse model of type II diabetes mellitus,and metformin hydrochloride(MET,200 mg/kg)was used in the positive group.The fasting blood glucose concentration,total cholesterol(TC),triglyceride(TG),low-density lipoprotein cholesterol(LDL-C),and high-density lipoprotein cholesterol(HDL-C)content in the serum were measured after gavage for three weeks in each group.The immunocompromised mouse model was constructed by intraperitoneal injection of cyclophosphamide(CTX,80 mg/kg)and divided into six groups the same as above,and the positive group was treated with levamisole(40 mg/kg).The immune organ index and cytokine content of the mice were measured after three weeks of gavage.The normal mice were grouped as above,and the positive group was treated with taurine(200 mg/kg).After four weeks of gavage,the weight-bearing swimming time of the mice was recorded,and blood urea nitrogen(BUN),lactic acid(LD)content,as well as activities of creatine kinase(CK),superoxide dismutase(SOD),and glutathione peroxidase(GSH-Px)were measured.The results showed that compared with the model group,the fasting blood glucose concentration,TC content,TG content,and LDL-C content of mice decreased to varying degrees after DP gavage,while the HDL-C content increased.In the immunomodulation experiment,the thymus and spleen index and cytokine content in the DP group showed a trend of increasing and were dose-dependent.The weight-bearing swimming time,GSH-Px activity,and SOD activity were significantly increased in t
