副猪嗜血杆菌PLGA-Omp16微球对小鼠树突状细胞免疫基因表达的影响

摘  要:将小鼠骨髓源树突状细胞(DCs)分别与副猪嗜血杆菌的Omp16蛋白、载Omp16的PLGA微球(PLGA-Omp16微球)以及LPS进行24 h共培养后,收集细胞提取RNA,用qPCR方法检测DCs细胞因子(IFN-γ、IL-1β、IL-6、TNF-α、INF-γ、CCL2和CXCL10)和表面标志物(MHC-II、CD80和CD86)mRNA的转录水平。结果显示,与各组抗原共培养24 h后,DCs细胞因子和表面标志物的mRNA转录水平显著高于PBS组(P<0.01);PLGA-Omp16微球组的TNF-α、IL-1β、CCL2、CXCL10、MHC-II、CD80和CD86等免疫分子mRNA转录水平显著高于Omp16组(P<0.05)。结果表明,PLGA-Omp16微球可增进DCs细胞因子转录水平,促进DCs的成熟。After co-culturing mouse bone marrow-derived dendritic cells(DCs)with the Omp 16 protein of Haemophilus parasuis,PLGA microspheres carrying Omp 16(PLGA-Omp 16 microspheres),and LPS for 24 hours,the cells were collected,and RNA was extracted.The mRNA transcription levels of DCs cytokines(IFN-γ,IL-1β,IL-6,TNF-α,INF-γ,CCL2,and CXCL10)and surface markers(MHC-II,CD80,and CD86)were detected by qPCR.The results reveal that after 24-hour coculture with each antigen group,the mRNA transcription levels of DCs cytokines and surface markers were significantly higher than those in the PBS group(P<0.01).The mRNA transcription levels of immune molecules such as TNF-α,IL-1β,CCL2,CXCL10,MHC-II,CD80,and CD86 in the PLGA-Omp 16 microsphere group were significantly higher than those in the Omp 16 group(P<0.05).The results show that PLGA-Omp16 microspheres could enhance the transcription level of DCs cytokines and promote the maturation of DCs.
