摘 要:采用十字交叉法和血球计数板法测试了温度、pH值、光照等培养条件和碳源、氮源等营养条件对红刺露兜病原尖孢镰刀菌菌丝生长与产孢的影响。结果表明,尖孢镰刀菌在10~40℃温度均能生长,适宜生长温度为20~30℃,最适生长温度为30℃;光照促进菌丝生长;菌落在pH值为4~12均能生长,最适pH值为8或9;不同碳氮源对尖孢镰刀菌的生长具有一定影响,最适宜碳氮源分别为蔗糖、硝酸钾。尖孢镰刀菌在15~40℃均能产孢,20~30℃适宜产孢,最适温度为30℃。光照促进产孢;pH值5~12均能产孢,最适产孢pH值为7;碳源培养基上产孢量普遍高于氮源培养基,最适碳氮源分别为山梨醇和牛肉膏。使用尿素、硫酸铵等不利于尖孢镰刀菌生长和产孢的叶面肥,可能有助于阻止尖孢镰刀菌侵入前期的定殖,从而降低红刺露兜褐斑病的发生率。The effects of culture conditions such as temperature,pH value,and light,as well as nutritional conditions such as carbon sources and nitrogen sources on the mycelial growth and sporulation of Fusarium oxysporum,the pathogen of Pandanus utilis,were tested by the cross method and the hemocytometer method.The results show that F.oxysporum could grow at temperatures ranging from 10 to 40℃.The suitable growth temperature was 20~30℃,and the optimum growth temperature was 30℃.Light promoted mycelial growth and the colony could grow at pH values ranging from 4 to 12,and the optimum pH value was 8 or 9.Different carbon and nitrogen sources had a certain influence on the growth of F.oxysporum.The most suitable carbon and nitrogen sources were sucrose and potassium nitrate respectively.F.oxysporum could sporulate at temperatures ranging from 15 to 40℃.The suitable sporulation temperature was 20~30℃,and the optimum temperature was 30℃.Light promoted sporulation.The fungus could sporulate at pH values ranging from 5 to 12,and the optimum sporulation pH value was 7.The sporulation amount on the carbon source medium was generally higher than that on the nitrogen source medium.The most suitable carbon and nitrogen sources were sorbitol and beef extract respectively.The use of foliar fertilizers such as urea and ammonium sulfate,which are not conducive to the growth and sporulation of F.oxysporum,may help prevent the invasion of F.oxysporum into the early stage of colonization,thereby reducing the incidence of brown spot disease of Pandanus utilis.
