摘 要:【目的】旨在探究长期高铜饲喂对绵羊胰腺组织损伤所产生的影响,从而为在饲料中科学合理地添加铜提供理论依据,为临床铜中毒提供分子病理学的研究基础。【方法】试验选用8只健康12月龄东北细毛羊母羊,随机分为2组,分别为4只饲喂正常日粮的对照组和4只在正常日粮基础上灌喂高铜(Cu)20 mg·kg^(-1)的试验组。高铜的灌喂时间为上午和下午各1次,于8周后通过鲍恩氏(Bouin)固定液、2.5%戊二醛或液氮冷冻等固定方法采集对照组和试验组的胰腺组织,利用病理切片苏木素-伊红(H.E.)染色和透射电子显微镜观察胰腺组织病理变化,测量胰腺腺泡直径和面积大小,免疫荧光和免疫组化检测胰腺组织天冬氨酸蛋白水解酶1(Caspase-1)、Gasdermins家族蛋白(DGSDMD)和白细胞介素-18(IL-18)等焦亡蛋白表达水平变化。【结果】与对照组相比高铜组胰腺组织明显水肿且腺泡直径和面积显著高于对照组(P<0.05);超微结构表明高铜组胰腺腺泡的内质网和线粒体发生损伤,符合细胞焦亡超微结构特征;与对照组相比高铜组胰腺组织Caspase-1、GSDMD和IL-18等焦亡标志性蛋白表达水平上升。【结论】长期高铜饲喂绵羊会导致绵羊胰腺组织通过细胞焦亡而发生损伤。[Objective]The aim is to explore the effects of long-term high-copper feeding on the histological damage of the pancreas in sheep,so as to provide a theoretical basis for the scientific and reasonable addition of copper in feed,and to provide a research basis of molecular pathology for clinical copper poisoning.[Methods]Eight healthy 12-month-old Northeast fine wool sheep were selected and randomly divided into two groups,including the control group in which four sheep were fed with normal diet,and the experimental group in which four sheep were fed with high copper(Cu 20 mg/kg)based on normal diet.The gavage time of high copper was once each in the morning and afternoon.After 8 weeks,sheep pancreatic tissues of the control and experimental groups were collected and put into Bouin fixative,2.5%glutaraldehyde,or liquid nitrogen.The pathological changes in pancreatic tissues were observed by transmission electron microscopy after hematoxylin-eosin(H.E.)staining of pathological sections,the diameter and area of the pancreatic acinar cells were measured,and the expression levels of pyroptosis proteins such as Caspase-1,GSDMD and IL-18 in pancreatic tissues were detected by immunofluorescence and immunohistochemistry methods.[Results]The results showed that compared with the control group,the pancreatic tissue of the experimental group swelled obviously and the diameter and area of the acinar cells were significantly larger.Ultrastructure analysis showed that the endoplasmic reticulum and mitochondria of pancreatic acinar cells in the experimental group were damaged,which was consistent with the ultrastructural characteristics of pyroptosis.The expression levels of Caspase-1,GSDMD and IL-18 in pancreatic tissue of the experimental group were higher than those of the control group.[Conclusion]In conclusion,high copper feeding to sheep would damage pancreatic tissue through pyroptosis.Data support was provided by this study for revealing the molecular mechanism of sheep pancreatic injury by high copper feeding,and theor
