摘  要:目的:探讨柴胡疏肝汤对抑郁大鼠抑郁样行为的改善作用及其机制。方法:本实验选取无特定病原体(SPF)级斯泼累格·多雷(SD)健康雄性大鼠,采取单次腹腔注射脂多糖(LPS)的方法建立抑郁大鼠模型,通过糖水偏好度测试和旷场实验评估大鼠的抑郁样行为。造模成功后,各组给予相应的方法进行干预,通过蛋白质免疫印迹法和实时聚合酶链反应(Real-time PCR)分别检测各组大鼠海马组织的多发性内分泌腺瘤1型基因编码蛋白(Menin)、胱天蛋白酶-3(Caspase-3)、核因子κB(NF-κB)、白细胞介素-1β(IL-1β)和mRNA表达水平,最终探讨柴胡疏肝汤对抑郁大鼠的影响机制。结果:单次腹腔注射LPS可诱导大鼠出现表情淡漠、双眼无神、食欲减退、活动减少等抑郁样行为。柴胡疏肝汤可显著恢复抑郁大鼠的体质量和运动能力(P<0.05,P<0.001)。与空白组比较,抑郁组大鼠的Menin表达水平显著降低(P<0.001),Caspase-3、NF-κB、IL-1β表达水平显著升高(P<0.001);与抑郁组比较,柴胡疏肝汤低剂量组、中剂量组、高剂量组Menin表达水平明显提高(P<0.05,P<0.01,P<0.001),NF-κB表达水平明显降低(P<0.05,P<0.001),Caspase-3、IL-1β表达水平显著降低(P<0.001)。结论:柴胡疏肝汤通过作用于Menin/Caspase-3信号通路,促进Menin表达,抑制Caspase-3表达,从而有效改善抑郁大鼠的抑郁样行为。Objective:To explore the effect of Chaihu Shugan Decoction on depressive-like behavior in rats and its underlying mechanisms.Methods:SPF-grade Sprague-Dawley(SD)male rats were selected for the experiment.A depressive rat model was established by a single intraperitoneal injection of lipopolysaccharide(LPS).Depressive-like behavior was assessed using the sucrose preference test and the open field test.After successful modeling,the rats in each group were treated with the corresponding interventions.Protein expression levels of multiple endocrine neoplasia type 1(Men1),cysteinyl aspartate-specific protease 3(Caspase-3),nuclear factor kappa B(NF-κB),interleukin-1β(IL-1β),and mRNA levels in the hippocampal tissue were measured using Western blot and real-time PCR,respectively.The mechanism by which Chaihu Shugan Decoction affected depressed rats was ultimately explored.Results:A single intraperitoneal injection of LPS induced depressive-like behavior in rats,including a dull expression,dull eyes,reduced appetite,and decreased activity.Chaihu Shugan Decoction significantly restored the body weight and motor ability of depressed rats(P<0.05,P<0.001).Compared to the blank group,the Menin expression level in the depression group was significantly reduced(P<0.001),while Caspase-3,NF-κB,and IL-1βexpression levels were significantly increased(P<0.001).Compared to the depression group,the low,medium,and high-dose groups of Chaihu Shugan Decoction significantly increased Menin expression levels(P<0.05,P<0.01,P<0.001),significantly decreased NF-κB expression levels(P<0.05,P<0.001),and significantly reduced Caspase-3 and IL-1βexpression levels(P<0.001).Conclusion:Chaihu Shugan Decoction improves depressive-like behavior in rats by regulating the Menin/Caspase-3 signaling pathway,promoting Menin expression,and inhibiting Caspase-3 expression.
