胃肠安抑制胃癌侵袭转移的机制研究

摘　　要：目的:探讨胃肠安通过趋化素样因子超家族6/程序性死亡受体配体1(CMTM6/PD-L1)轴抑制胃癌侵袭转移的分子机制。方法:选择人胃癌MKN45、MGC803细胞,细胞增殖实验检测胃肠安对细胞增殖的抑制能力;划痕实验和侵袭实验检测胃肠安对细胞迁移和侵袭的抑制能力;实时荧光定量核酸扩增检测各组CMTM6、PD-L1信使核糖核酸(mRNA)水平;免疫荧光实验检测各组CMTM6、PD-L1蛋白荧光强度;蛋白质免疫印迹实验检测各组基质金属蛋白酶2(MMP2)、基质金属蛋白酶9(MMP9)、上皮型钙黏蛋白(E-cadherin)、神经型钙黏蛋白(N-cadherin)、波形蛋白(Vimentin)、CMTM6、PD-L1蛋白表达水平。结果:胃肠安可以抑制人胃癌MKN45、MGC803细胞的增殖、迁移和侵袭能力,降低MMP2、MMP9、N-cadherin、Vimentin蛋白表达量,升高E-cadherin蛋白表达量,降低CMTM6、PD-L1蛋白和mRNA表达量。结论:胃肠安可以抑制人胃癌MKN45、MGC803细胞的迁移、侵袭及上皮-间充质转化,其作用机制可能与CMTM6/PD-L1轴有关。Objective:To explore the molecular mechanism by which Weichang′an inhibits the invasion and metastasis of gastric cancer through the chemokine-like factor(CKLF)-like MARVEL transmembrane domain-containing protein 6(CMTM6)/programmed death ligand 1(PD-L1)axis.Methods:Human gastric cancer cell lines MKN45 and MGC803 were used.The cell proliferation was assessed by a cell proliferation assay to determine the inhibitory effect of Weichang′an on cell growth.The inhibitory effect of Weichang′an on cell migration and invasion were measured using scratch assay and invasion assay,respectively.The mRNA levels of CMTM6 and PD-L1 in each group were quantified by real-time fluorescence quantitative PCR.Immunofluorescence staining was used to detect the protein fluorescence intensity of CMTM6 and PD-L1.Protein levels of MMP2,MMP9,E-cadherin,N-cadherin,vimentin,CMTM6,and PD-L1 were measured using Western blot.Results:Weichang′an significantly inhibited the proliferation,migration,and invasion of human gastric cancer cells(MKN45 and MGC803).It decreased the expression levels of MMP2,MMP9,N-cadherin,and vimentin,while increasing the expression of E-cadherin.Additionally,it reduced both the protein and mRNA expression levels of CMTM6 and PD-L1.Conclusion:Weichang′an can inhibit the migration,invasion,and epithelial-mesenchymal transition(EMT)of human gastric cancer cells,and its mechanism may be related to the regulation of the CMTM6/PD-L1 axis.
