摘 要:目的:研究人参固本口服液醇沉药渣中多糖的组成及活性,实现中药资源综合利用。方法:采用高效液相色谱法(HPLC)分析单糖组成;采用盐酸林可霉素构建的肠道微生态失调雄性Balb/c小鼠模型研究微生态调节作用,实验设自然恢复组、丽珠肠乐组(0.5亿活菌/mL的丽珠肠乐溶液)、多糖低剂量组(75 mg/kg)和多糖高剂量组(150 mg/kg)。采用刀豆蛋白A(ConA)诱导的小鼠脾淋巴细胞转化实验、抗体生成细胞检测和自然杀伤细胞(NK)活性测定考察对小鼠免疫功能的影响,实验设阴性对照组、多糖低剂量组(75 mg/kg)和多糖高剂量组(150 mg/kg)。结果:人参固本口服液醇沉药渣中多糖含量达68.5%,组成包括鼠李糖、阿拉伯糖、半乳糖、半乳糖醛酸、无水葡萄糖、葡萄糖醛酸、甘露糖、木糖、岩藻糖和核糖10种单糖。与自然恢复组比较,多糖低、高剂量组均能显著改善小鼠肠道菌群失调状况(P<0.05),且多糖高剂量组改善效果优于丽珠肠乐组(P<0.05)。与阴性对照组比较,多糖高剂量组可显著促进淋巴细胞增殖(P<0.05),增加抗体生成细胞数量(P<0.05),增强NK细胞活性(P<0.01)。结论:人参固本口服液醇沉药渣中多糖主要由无水葡萄糖、半乳糖等10种单糖组成。药渣多糖能调节小鼠肠道微生态并提高免疫力,具有重要的开发利用价值。Objective:To study the composition and activity of polysaccharides in the alcohol precipitate from Renshen Guben Oral Liquid,with the aim of achieving comprehensive utilization of Chinese medicinal resources.Methods:High-performance liquid chromatography(HPLC)was used to analyze the monosaccharide composition.An intestinal microecology imbalance model in male Balb/c mice was constructed using lincomycin hydrochloride to study the microecological regulation effect.The experimental groups included the natural recovery group,a bifidobacterium(Lizhu Changle)group(0.5×109 CFU/mL of Lizhu Changle solution),low-dose polysaccharide group(75 mg/kg),and high-dose polysaccharide group(150 mg/kg).The effect on immune function was assessed through a series of experiments,such as spleen lymphocyte transformation assay using ConA,antibody-forming cell detection,and natural killer(NK)cell activity measurement.Negative control group,low-dose polysaccharide group(75 mg/kg),and high-dose polysaccharide group(150 mg/kg)were set up.Results:The polysaccharide content in the alcohol precipitate of Renshen Guben Oral Liquid was 68.5%,and the composition included ten monosaccharides,including rhamnose,arabinose,galactose,galacturonic acid,anhydrous glucose,glucuronic acid,mannose,xylose,fucose,and ribose.Compared with the natural recovery group,both the low-dose and high-dose polysaccharide groups significantly improved the intestinal microbiota imbalance in mice(P<0.05),with the high-dose polysaccharide group showing better effects than the Lizhu Changle group(P<0.05).Compared with the negative control group,the high-dose polysaccharide group significantly promoted lymphocyte proliferation(P<0.05),increased the number of antibody-forming cells(P<0.05),and enhanced NK cell activity(P<0.01).Conclusion:The polysaccharides in the alcohol precipitate of Renshen Guben Oral Liquid are primarily composed of ten monosaccharides,including anhydrous glucose and galactose.The polysaccharides from the residue can regulate the intestinal microecolog
