摘  要:目的探讨LncRNA-ZMIZ1-AS1在原发性肝癌(HCC)中调控细胞生长、侵袭及生物学行为的具体作用机制。方法本研究通过生物信息学分析结合GEO和TCGA数据,分析LncRNA-ZMIZ1-AS1与HCC临床预后的关系。利用实时PCR、蛋白质印迹等分子生物学实验,以及细胞活力分析、流式细胞术等技术,探讨敲低LncRNA-ZMIZ1-AS1对HCC细胞增殖、迁移、侵袭和凋亡的影响。结果LncRNA-ZMIZ1-AS1在HCC患者中高表达,且与性别、临床分期等病理特征显著相关。敲低LncRNA-ZMIZ1-AS1显著抑制HCC细胞的增殖、迁移和侵袭能力,并增加细胞凋亡率。此外,GSEA分析表明,LncRNAZMIZ1-AS1的高表达可能与NOTCH信号通路相关联。结论LncRNA-ZMIZ1-AS1在HCC中的高表达可能通过调控NOTCH等信号通路,促进肿瘤细胞的恶性表型。其在HCC中的表达具有潜在的生物标志物意义,为进一步研究其在肝癌中的分子机制及其在临床中的应用潜力提供了理论依据。Objective To investigate the specific mechanisms of long non-coding RNA ZMIZ1-AS1(LncRNA-ZMIZ1-AS1)regulating cell growth,invasion and biological behavior in hepatocellular carcinoma(HCC).Methods This study used bioinformatics analysis combined with GEO and TCGA data to analyze the relationship between LncRNA-ZMIZ1-AS1 and the clinical prognosis of HCC patients.Molecular biology experiments such as real-time PCR and Western blotting as well as cell viability analysis,flow cytometry and other technologies were used to explore the effects of knocking down LncRNA-ZMIZ1-AS1 on HCC cell proliferation,migration,invasion and apoptosis.Results LncRNA-ZMIZ1-AS1 was highly expressed in HCC patients.It was significantly associated with clinical characteristics such as gender and clinical stage.Knockdown of LncRNA-ZMIZ1-AS1 notably inhibited HCC cell proliferation,migration and invasion.It also increased apoptosis rates.Additionally,GSEA analysis showed a potential association between high LncRNA-ZMIZ1-AS1 expression and the NOTCH signaling pathway.Conclusions The high expression of LncRNA-ZMIZ1-AS1 in HCC likely promotes malignant phenotypes by regulating signaling pathways such as NOTCH.This finding underscores its potential as a biomarker for HCC.It provides a theoretical basis for further research on its molecular mechanisms and clinical applications in liver cancer.
