摘 要:目的 本研究旨在调查PTX3基因在胶质瘤中的表达及其与患者预后的关联。方法 利用癌症基因组图谱(TCGA)和基因型-组织表达(GTEx)数据库的数据,全面分析:差异基因表达、功能和通路富集、免疫浸润分析和统计评估。进行涵盖33种癌症类型的单变量Cox回归分析,评估PTX3的预后意义。结果 PTX3在低级别胶质瘤和胶质母细胞瘤中的表达显著升高,与患者预后不良相关。此外,通过基因本体(GO)和京都基因与基因组百科全书(KEGG)的富集分析突出了PTX3在细胞因子介导的信号传导、细胞迁移和免疫反应通路中的参与。值得注意的是,PTX3的表达与巨噬细胞和Th2细胞的浸润呈正相关,而与CD8 T细胞和调节性T细胞呈负相关,表明其在调节胶质瘤免疫微环境中的关键作用。此外,PTX3水平随着肿瘤进展而增加,与更高的WHO分级、IDH野生型状态和患者年龄呈正相关。结论 PTX3是胶质瘤预后的一种有前景的生物标志物,但需要进一步研究以阐明其功能的分子机制,并探索其在胶质瘤治疗中的临床应用。Objective This study aims to investigate PTX3 gene expression in glioblastoma and its association with patient prognosis.Methods Using data from The Cancer Genome Atlas(TCGA) and Genotype-Tissue Expression(GTEx) databases,we performed a comprehensive analysis encompassing differential gene expression,functional and pathway enrichment,immune infiltration analysis,and statistical evaluation.A uni-variate Cox regression analysis covering 33 cancer types was conducted to assess the prognostic significance of PTX3.Results Our findings reveal that PTX3 expression is significantly elevated in low-grade gliomas and glioblastomas,correlating with poor patient prognosis.Furthermore,enrichment analyses using Gene Ontology(GO) and the Kyoto Encyclopedia of Genes and Genomes(KEGG) highlighted PTX3's involvement in cytokine-mediated signaling,cell migration,and immune response pathways.Notably,PTX3 expression exhibited a positive correlation with the infiltration of macrophages and Th2 cells,while showing a negative correlation with CD8+ T cells and regulatory T cells,underscoring its critical role in modulating the glioma immune microenvironment.Additionally,PTX3 levels were found to increase with tumor progression,positively associated with higher WHO grades,IDH wild-type status,and patient age.Conclusion Our study highlights PTX3 as a promising prognostic biomarker for gliomas.Further research is required to elucidate its molecular mechanisms and explore its clinical applications in glioma treatment.
