摘 要:目的:探讨八珍汤(Bazhen decoction)对自然衰老果蝇的改善作用。方法:采用爬管实验,计算攀爬指数,分析八珍汤对果蝇运动能力的影响;通过记录果蝇产卵数量,分析八珍汤对果蝇生殖能力的影响;通过绘制果蝇自然死亡生存曲线,分析八珍汤对果蝇寿命的影响;利用酶联免疫吸附实验,检测果蝇体内氧化应激相关生化指标,包括超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和活性氧(ROS),以评估八珍汤的抗氧化功能。结果:0.1 g/mL浓度的八珍汤显著延长果蝇的寿命(P<0.001),平均生存期、中位生存期、最高生存期以及延寿率均有显著提升;此外,八珍汤还能够提高果蝇的运动能力(P<0.05)及繁殖能力(P<0.01),并显著提高果蝇体内SOD活力、CAT水平,降低ROS含量(P<0.05)。结论:八珍汤可通过抗氧化应激、调节氧化还原发挥抗衰老功效。Objective:To investigate the improvement effect of Bazhen decoction on naturally aging drosophila melanogaster.Methods:The climbing tube test was used to calculate the climbing index and analyze the effect of Bazhen decoction on the motor ability of drosophila melanogaster.The egg-laying number of drosophila melanogaster was recorded to analyze the effect of Bazhen decoction on their reproductive ability.The natural death survival curve of drosophila melanogaster was plotted to analyze the effect of Bazhen decoction on their lifespan.Enzyme-linked immunosorbent assay(ELISA)was used to detect oxidative stress-related biochemical indicators in drosophila melanogaster,including superoxide dismutase(SOD),catalase(CAT)and reactive oxygen species(ROS),to evaluate the antioxidant function of Bazhen decoction.Results:Bazhen Decoction at a concentration of 0.1 g/mL significantly prolonged the lifespan of drosophila melanogaster(P<0.001),with significant improvements in mean survival time,median survival time,maximum survival time and life extension rate.Additionally,Bazhen decoction improved the motor ability(P<0.05)and reproductive ability(P<0.01)of drosophila melanogaster,and significantly increased SOD activity and CAT levels while decreasing ROS content in drosophila melanogaster(P<0.05).Conclusion:Bazhen decoction exerts anti-aging effects by anti-oxidative stress and regulating redox status.
