摘  要:目的探讨18β-甘草次酸(18β-glycyrrhetinic acid,18β-gly)对失血性休克大鼠肺组织损伤及炎症因子TNF-α、IL-1β的影响。方法实验用SD雌性大鼠随机分为5组:假手术组、模型组、18β-Gly/低剂量组、18β-Gly/中剂量组、18β-Gly/高剂量组,通过18β-甘草次酸预处理,采用股动脉放血制作失血性休克大鼠模型,检测大鼠失血性休克后肺组织湿/干比、肺组织病理形态变化、血液及肺泡灌洗液中炎症因子肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)的表达情况。结果与模型组相比,18β-Gly/高剂量组肺湿/干比(4.409±0.062)降低比较明显(P<0.05);血液及肺泡灌洗液中18β-Gly/中剂量组和18β-Gly/高剂量组,TNF-α和IL-1β的表达均明显减少(P<0.05)。HE显示,与模型组相比,18β-Gly/中剂量组和18β-Gly/高剂量组病理损伤明显减轻。结论18β-Gly能减轻失血性休克大鼠肺水肿和肺组织病理损伤,降低血液及肺泡灌洗液中炎症因子TNF-α、IL-1β的表达。Objective To investigate the effects of 18β-glycyrrhetinic acid(18β-glycyrrhetinic acid,18β-gly)on lung injury and inflammatory cytokines TNF-αand IL-1βin hemorrhagic shock rats.Methods SD Female rats were randomLy divided into 5 groups:sham operation group,model group,18β-Gly/low dose group,18β-Gly/medium dose group and 18β-Gly/high dose group.After 18β-glycyrhetinic acid pretreatment,the rat model of hemorhagic shock was made by femoral artery bloodlet.The wet/dry ratio of lung tissue,pathological morphological changes of lung tissue,and expression of inflammatory factors TNF-αand IL-1βin blood and alveolar lavage fluid were detected.Results Compared with model group,the lung wet/dry ratio in 18β-Gly/high-dose group(4.409±0.062)was significantly decreased(P<0.05).The expressions of TNF-αand IL-1βin blood and alveolar lavage fluid of 18β-Gly/medium dose group and 18β-Gly/high dose group were significantly decreased(P<0.05).HE showed that pathological damage was significantly reduced in the 18β-Gly/medium dose and 18β-Gly/high dose groups compared with the model group.Conclusion 18β-Gly can alleviate pulmonary edema and pathological damage of lung tissue in hemorrhagic shock rats,and decrease the expression of inflammatory cytokines TNF-αand IL-1βin blood and alveolar lavage fluid.
