摘 要:DNA除了具有典型的右手双螺旋结构外,还具有多种多样的二级结构。i-Motif DNA是富含胞嘧啶碱基(C)的序列,以半质子化的C·C^(+)碱基对为基础形成的特殊二级结构。其广泛存在于染色体端粒末端和基因启动子区域,并与多种癌症的发生相关,成为新型的癌症治疗靶点。此外,因i-motif结构具有独特的pH敏感性,可作为功能核酸的基本结构单元,在纳米生物技术与疾病诊疗方面具有重要应用。本文首先就i-motif的结构性质和稳定性,以及小分子配体进行总结,随后就i-motif在生物传感以及药物释放方面应用进行阐述。Apart from the typical right-handed double helix structure,DNA also features a variety of secondary structures.One such structure is the i-motif DNA,which is formed by cytosine-rich sequences with semi-protonated C·C^(+)base pairs.The presence of i-motif in telomeres and cancer gene promoters has been confirmed,making it a new target for cancer treatment.Additionally,i-motif can serve as a fundamental structural unit in functional nucleic acids due to its pH sensitivity.i-Motif-based DNA bionanotechnology has been utilized in diagnosis and treatment.This review will summarize the structural properties and stability of i-motif,as well as the small-molecular ligands targeting i-motif.Furthermore,it will discuss the applications of i-motif in biosensing and drug release.
