摘 要:目的:研究金钗石斛和玫瑰石斛茎的低极性成分及抗肿瘤活性。方法:经95%乙醇提取,石油醚萃取,对两种石斛属植物茎中低极性成分进行GC-MS分析。以MTS法检测肿瘤细胞毒活性,筛选其乙醇提取物、石油醚萃取物的抗肿瘤活性。结果:金钗石斛茎低极性成分GC-MS检出匹配度大于85%的化合物有16种,相对含量占94.06%。其茎石油醚萃取物浓度在200μg/mL对白血病(HL-60)和结肠癌(SW480)两种肿瘤细胞抑制率分别为89.02%和70.73%。玫瑰石斛茎低极性成分GC-MS检出匹配度大于85%的化合物有3种,相对含量占88.87%。其茎乙醇萃取物浓度在200μg/mL对白血病(HL-60)、乳腺癌(MDA-MB-231)、结肠癌(SW480)3种肿瘤细胞抑制率分别为92.47%、89.67%和91.10%。结论:通过两种植物茎低极性成分的GC-MS分析及抗肿瘤活性研究,丰富了其化学成分的研究,进一步验证了其抗肿瘤活性。Objective:To study the low polarity components in the stem of Dendrobium nobile and Dendrobium crepidatum,as well as the anti-tumor activity of its’ethanol extracts and petroleum ether extracts.Methods:The stem of D.nobile and D.crepidatum were extracted with ethanol and were identified by GC-MS respectively.The relative contents of the chemical constituents were determined by area normalization method.Detect the cytotoxicity of tumor cells using MTS method and screen the anti-tumor activity of its ethanol extract and petroleum ether extract.Results:16 compounds with a matching degree greater than 85%detected by GC-MS for the low polarity components in the stem of D.nobile with a relative content of 94.06%.The inhibitory rates of its stem petroleum ether extract at a concentration of 200μg/mL on leukemia(HL-60)and colon cancer(SW480)tumor cells were 89.02%and 70.73%,respectively.32 compounds with a matching degree greater than 85%detected by GC-MS for the low polarity components in the stem of D.crepidatum with a relative content of 88.87%.The inhibitory rates of its stem petroleum ether extract at a concentration of 200μg/mL leukemia(HL-60),breast cancer(MDA-MB-231)and colon cancer(SW480)was 92.47%,89.67%and 91.10%,respectively.Conclusion:Through GC-MS analysis and anti-tumor activity research on the low polarity components of these two plants,the research on its chemical components has been enriched,and its anti-tumor activity has been further verified.
