摘  要:目的:优选黔产白及经“蒸制”加工与炮制一体化工艺。方法:用不同蒸制加工工艺处理新鲜白及,提取白及多糖(bletilla striata polysaccharide,BSP)进行抗炎和凝血、止血作用药理实验研究并作为优选指标。结果:在断尾实验中,与空白组比较,白及多糖组和云南白药组出血时间均缩短,其中以BSP9组效果最明显;毛细玻管实验中,与空白组比较,白及多糖组和云南白药组的凝血时间缩短,以BSP8、BSP9、BSP11组明显缩短。角叉菜胶致小鼠足肿胀实验结果表明,与模型组相比,在4 h时,白及多糖组都能抑制足肿胀程度,尤以BSP6、BSP7、BSP9组抑制效果最显著。二甲苯致小鼠耳廓肿胀结果显示,与模型组比较,除BSP6组,其他组均能减轻肿胀程度,其中以BSP7、BSP9、BSP11组减轻效果最好;通过Elisa结果显示,与模型组相比,各白及多糖组的小鼠血清中肿瘤坏死因子α(TNF-α)、白细胞介素6(IL-6)、前列腺素E2(PEG2)的表达降低,其中以BSP6、BSP7、BSP8、BSP11组显著抑制炎症因子表达。结论:优选的最佳一体化工艺为BSP9组:洗净,不去须根,100℃蒸8 min,切片,去须根,烘干,碱提纯沉法提多糖。Objective To optimize the processing and processing technology of bletilla striata in Guizhou.Methods Bletilla striata polysaccharide(BSP)was extracted and treated with different steaming processes to study its anti-inflammatory,coagulation and hemostatic effects.Results In the tail breaking experiment,compared with the blank group,the bleeding time in both the bletilla striata polysaccharide group and Yunnan Baiyao group was shortened,and the effect of BSP9 group was the most obvious.In capillary tube experiment,compared with the blank group,the coagulation time of Bletilla striata polysaccharide group and Yunnan Baiyao group was shortened,and the coagulation time of BSP8,BSP9 and BSP11 groups was significantly shortened.The experimental results of foot swelling induced by carrageenan showed that,compared with model group,both bletilla striata polysaccharide groups could inhibit foot swelling at 4 h,especially BSP6,BSP7 and BSP9 groups.The results of ear swelling induced by xylene showed that compared with model group,except BSP6 group,other groups could reduce the degree of swelling,and BSP7,9 and 11 groups had the best effect.Elisa results showed that compared with the model group,the expressions of TNF-α,IL-6 and PEG2 in serum of mice in the bletilla striata polysaccharide groups were decreased,and the expressions of inflammatory factors in BSP6,BSP7,BSP8 and 11 groups were significantly inhibited.Conclusion The optimal integrated process is as follows BSP9 group:wash,no root removal,steaming at 100℃for 8 min,slicing,root removal,drying,alkali purification and precipitation extraction of polysaccharide.
