摘 要:目的:优选玛努-4颗粒水提取工艺及制备工艺条件。方法:采用L_(9)(3)^(4)正交试验,以异土木香内酯与浸膏提取率为评分标准,考察指标为浸泡时间、提取时间、加水量、提取次数,综合评分优选优选水提取工艺;用玛努-4浸膏为原料,考察指标为成型率、吸湿率、堆密度、溶化性、休止角,对乙醇浓度、乙醇用量、辅料选择和药粉与辅料的比例进行单因素考察,综合评分优选优选成型工艺。结果:优选水提取工艺为浸泡90 min、加16倍量水、煎煮1 h、提取1次;优选颗粒成型工艺为润湿剂选用80%乙醇、浸膏与润湿剂比例为5∶1、浸膏与乳糖比例为3∶2制得的颗粒成型性较好。结论:提取工艺便捷可靠,能高效提取有效成分;颗粒剂成型工艺稳定可控,为玛努-4颗粒的开发应用提供实验依据。Objective Optimize selection of water extraction process and preparation conditions for Manu-4 particles.Methods L_(9)(3)^(4)orthogonal experiment was used to select the extraction rate of codenolactone and extract as the scoring standard.The indexes were soaking time,extraction time,amount of water added and extraction times.Extractum Manu-4 was used as raw material,and the indexes were molding rate,hygrometric rate,bulk density,solubility and resting Angle.The ethanol concentration,ethanol dosage,selection of auxiliary materials and the ratio of powder to auxiliary materials were investigated by single factor.Results The optimal water extraction process was soaked for 90 min,added 16 times of water,decocted for 1h,and extracted once.The optimum particle forming process was 80%ethanol as wetting agent,5∶1 ratio of extractum to wetting agent,3∶2 ratio of extractum to lactose.Conclusion:The extraction process is convenient and reliable,and can extract effective components efficiently.The molding process of the granules was stable and controllable,which provided experimental basis for the development and application of Manu-4 Granule.
