摘  要:目的:建立水田七中根薯酮内酯A和根薯酮内酯B的液质联用检测(LC-MS)和分析方法。方法:采用Kromasil Proshell C_(18)柱(2.5μm,2.1 mm×100 mm)色谱柱,以乙腈-0.1%甲酸溶液为流动相,梯度洗脱,多反应监测模式进行检测分析。结果:根薯酮内酯A和根薯酮内酯B进样量分别在10.1136~505.6800 pg和0.9996~99.9600 pg范围内线性关系良好,r>0.999,加样回收率分别为95.45%(RSD 2.88%),99.12%(RSD 2.82%)。结论:所建立的方法快速、准确、灵敏、可靠,能够用于水田七中根薯酮内酯A和根薯酮内酯B的测定,为其质量控制研究提供科学依据。Objective To establish the analysis method of taccalonolide A and taccalonolide B in Schizocapsa Plantaginea Rhizoma by liquid chromatography-mass spectrometry(LC-MS).Methods An Kromasil Proshell C_(18)(2.5μm,2.1 mm×100 mm)column was used with gradient elution of acetonitrile-0.1%formic acid.The multiple-reaction monitoring mode was used.As a result,the LC-MS method was established to detect the taccalonolide A and taccalonolideB in Schizocapsa Plantaginea Rhizoma.And the method was further validated and used to test all samples.Results The developed method showed good linearity in the range of 10.1136-505.6800 pg,0.9996-99.9600 pg with r>0.999 for taccalonolide A and taccalonolide B,respectively.The recovery rates were 95.45%(RSD 2.88%),99.12%(RSD 2.82%),respectively.Conclusion The established method is accurate,sensitive and reliable.It can be applied for analysis of taccalonolide A and taccalonolide B in Schizocapsa Plantaginea Rhizoma.And it can provide sound scientific basis for the study of Schizocapsa plantaginea rhizome quality control.
