摘 要:目的优化青蒿抗炎有效部位的提取工艺,并评价其抗炎活性。方法采用正交实验设计考察料液比、乙醇体积分数、提取温度和提取时间对从青蒿中提取抗炎有效部位的影响,确定最佳提取工艺参数;建立脂多糖(lipopolysaccharide,LPS)刺激的RAW 264.7细胞炎症模型,通过测定一氧化氮(nitric oxide,NO)含量体外评价青蒿提取物的抗炎活性;通过建立二甲苯致小鼠耳肿胀模型体内验证青蒿提取物的抗炎作用。结果从青蒿中提取抗炎有效部位的条件是料液比为1∶5、乙醇体积分数为80%、提取温度为70℃、提取时间为2 h。青蒿提取物能够显著降低RAW 264.7炎症细胞的NO、肿瘤坏死因子-α(tumor necrosis factor,TNF-α)和白细胞介素-6(interleukin-6,IL-6)水平,同时可减轻二甲苯致小鼠耳肿胀的程度。结论确定了从青蒿中获得最佳抗炎提取物的制备工艺,为拓宽青蒿药材的药效应用提供科学依据。Objective To optimize the extraction process of anti-inflammatory components of Artemisia caruifolia Buch.-Ham.ex Roxb.and to evaluate their anti-inflammatory activity.Methods First,orthogonal experiments were conducted to investigate the effects of solid-liquid ratio,ethanol concentration,extraction temperature and extraction time on the preparation of anti-inflammatory extracts,and thereby confirming optimal parameters of the extraction process.Then,inflammatory model was established on RAW 264.7 cell with lipopolysaccharide(LPS).Nitric oxide(NO)content was measured to evaluate the anti-inflammatory effect of the extract from Artemisia caruifolia.Finally,the anti-inflammatory activity was in-vivo validated on the mouse model of ear swelling induced by xylene.Results The optimal extraction parameters of anti-inflammatory effective part from Artemisia caruifolia were determined as follows:A solid-liquid ratio of 1∶15,an ethanol concentration of 80%,an extraction temperature of 70℃and an extraction time of 2 hours.The active components from this plant with this extraction process can significantly reduce the concentrations of NO,tumor necrosis factor-α(TNF-α)and interleukin-6(IL-6)in RAW 264.7 inflammatory cells,and reduce the degree of ear swelling induced by xylene in mice.Conclusion This study established the preparation procedure for the optimally antiinflammatory extract from Artemisia caruifolia,providing a scientific basis for expanding the medicinal application of this plant.
