摘 要:目的建立九子不离母的质量控制方法,确保用药准确、质量可控,促进九子不离母的可持续发展。方法参照《云南省中药材(民族药材)质量标准研究技术指导原则》中的要求,采用基源鉴定、性状鉴定、显微鉴别、薄层色谱及高效液相色谱等方法,对九子不离母的原植物形态、性状特征、显微特征、薄层色谱进行研究,对水分、总灰分、酸不溶性灰分和醇浸出物含量、薯蓣皂苷含量进行测定。结果九子不离母药材基源为叉蕊薯蓣,性状、显微特征及薄层色谱鉴别明显。10批样品中,水分为6.88%~8.61%,总灰分为3.16%~4.62%,酸不溶性灰分为0.21%~0.76%,醇溶性浸出物含量为22.3%~33.1%,薯蓣皂苷含量为1.56%~2.02%。限度为水分不得超过10.0%,总灰分不得超过5.0%,酸不溶性灰分不得超过0.7%,水溶性浸出物不得少于20%,含薯蓣皂苷按干燥品计不得少于1.4%。结论九子不离母标准中的性状、显微特征明显,薄层色谱分离效果较好,水分、总灰分、酸不溶性灰分、醇浸出物测定方法可行,薯蓣皂苷含量测定方法简便、准确,重复性良好,为九子不离母的质量控制提供依据。Objective To establish a quality control method for Dioscorea collettii Hook.f.,ensure accurate medication and controllable quality,and promote the sustainable development of Dioscorea collettii Hook.f.Method In accordance with the requirements of the Technical Guidelines for Research on Quality Standards of Traditional Chinese Medicine(Ethnic Medicinal Materials)in Yunnan Province,methods such as original identification,trait identification,microscopic identification,thin-layer chromatography identification,and high-performance liquid chromatography(HPLC)were used to study the original plant morphology,trait characteristics,microscopic characteristics,and thin-layer chromatography(TLC)of Dioscorea collettii Hook.f.Water content,total ash content,acid insoluble ash content,alcohol extract content,and dioscin content were determined.Result The base source of Dioscorea collettii Hook.f.,was Dioscorea opposita,with obvious characteristics,microscopic features,and TLC spots.In the 10 batches of samples,the moisture content was 6.88%—8.61%,the total ash content was 3.16%—4.62%,the acid insoluble ash content was 0.21%—0.76%,the alcohol soluble extract content was 22.3%—33.1%,and the dioscin content was 1.56%—2.02%.The limit is that the moisture content≤10.0%,the total ash content≤5.0%,the acid insoluble ash content≤0.7%,the water-soluble extract≥20%,and the dry content of dioscin≥1.4%.Conclusion The characteristics and microscopic features in the standard of Dioscorea collettii Hook.f.are obvious,and the TLC separation has good effect.The determination method of moisture,total ash,acid insoluble ash,and alcohol extract is feasible.The determination method of dioscin content is simple,accurate,and has good repeatability,providing a basis for the quality control of Dioscorea collettii Hook.f.
