摘 要:目的通过网络药理学和动物实验探讨避瘟解毒颗粒(Biwen Jiedu Granules,BWJD)防治新型冠状病毒感染的机制。方法通过TCMSP和BATMAN-TCM数据库检索组方药材的化学成分及作用靶点。用GeneCards数据库查找新型冠状病毒感染相关靶点,将二者交集靶点导入STRING数据库中构建靶点相互作用网络图,筛选核心靶点,通过DVIAD数据库对靶点进行GO功能分析和KEGG信号通络富集分析。构建环磷酰胺诱导的小鼠免疫低下模型,检测小鼠的脾脏和胸腺指数、血常规、血清及组织中免疫因子水平。结果获得BWJD药物靶点259个,疾病靶点2284个,交集靶点98个,通过蛋白相互作用(proteinprotein interaction,PPI)网络分析获得肿瘤坏死因子(tumor necrosis factor,TNF)、白细胞介素-1β(interleukin-1β,IL-1β)、白细胞介素-6(interleukin-6,IL-6)等核心靶点。基因GO功能、KEGG通路富集分析结果表明,药物增强免疫功能主要是通过TNF、白细胞介素-17(interleukin-17,IL-17)等信号通路发挥协同调节作用。动物实验进一步证实BWJD能显著改善免疫低下小鼠的脾脏、胸腺指数和血常规,显著提高血清中免疫球蛋白G(immunoglobulin G,IgG)、免疫球蛋白M(immunoglobulin M,IgM)、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、IL-1β水平和脾、胸腺中TNF-α、IL-1β的含量,提高脾和胸腺中超氧化物歧化酶(superoxide dismutase,SOD)的活性。结论避瘟解毒颗粒可能通过免疫调节发挥防治新型冠状病毒感染的作用。Objective To explore the mechanism of Biwen Jiedu Granules(BWJD)in preventing and treating novel coronavirus infection based on network pharmacology and animal experiments.Methods The active constituents and targets of BWJD were retrieved through Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP)and A Bioinformatics Analysis Tool for Molecular Mechanism of Traditional Chinese Medicine(BATMAN-TCM)databases.Targets of novel coronavirus infection were searched from the GeneCards database,and their overlapping targets were imported into the STRING database to establish the interaction network.Furthermore,the core targets were screened out.Gene ontology(GO)function and Kyoto encyclopedia of genes and genomes(KEGG)pathway enrichment analysis were performed through the DVIAD database.An immunosuppression model of mice induced by cyclophosphamide was established,and the spleen and thymus index,hemogram,serum and tissue levels of immune factors were detected.Results A total of 259 drug targets,2284 disease targets and 98 overlapping targets were obtained for BWJD.Key targets such as tumor necrosis factor(TNF),interleukin-1β(IL-1β)and interleukin-6(IL-6)were performed through protein-protein interaction(PPI)network analysis.GO and KEGG pathway enrichment analysis revealed that BWJD could enhance immune function,mainly through synergistic regulatory effects such as TNF and IL-17 signaling pathway.Animal experiments further confirmed that BWJD could significantly improve the spleen,thymus index,and blood count levels in immunocompromised mice,and significantly increase the contents of immunoglobulin G(IgG),immunoglobulin M(IgM),tumor necrosis factor-α(TNF-α)and IL-1β.The contents of TNF,IL-1βand the activity of superoxide dismutase(SOD)in the spleen and thymus were significantly increased.Conclusion BWJD may play a role in preventing and treating novel coronavirus infection through immune regulation.
