摘 要:牦牛是青藏高原地区的特有动物之一,但其繁殖率一直较低,围产后期生殖系统恢复不及时是主要原因之一。本研究通过蛋白质组学技术筛选牦牛围产后期影响其产后恢复的关键血液蛋白,旨在探究牦牛产后生殖系统不能及时恢复的发生机制。试验选择青海地区经产牦牛40头,自分娩日起,每7天采集血液样本,根据时间点分为5组:A组(分娩日)、B组(产后7 d)、C组(产后14 d)、D组(产后21 d)和E组(产后28 d)。基于数据非依赖型扫描模式的蛋白质组学技术,筛选各组的蛋白质谱,并进行两两对比。结果获得718个鉴定的蛋白质,其中在任意两组中差异表达的有399种;对差异表达蛋白进行基因功能分析和信号通路分析后,筛选出与牦牛卵巢生殖激素分泌、卵巢糖代谢和脂类代谢相关的18种关键蛋白,并绘制了牦牛产后信号通路。通过对关键差异蛋白质进行分析,发现牦牛产后卵巢内生殖激素分泌和代谢的异常,以及糖代谢和脂类代谢供能的异常是导致牦牛产后生殖系统不能及时恢复的主要原因。Yak is one of the endemic animals in the Qinghai Tibet Plateau region,but its reproductive rate is consistently low,and the delayed recovery of their reproductive system during the late perinatal period is one of the main reasons.This study screened key proteins that affect postpartum recovery in yak during the late perinatal period using proteomics techniques,aiming to explore the mechanism of delayed postpartum recovery in yak during the late perinatal period.Forty multiparous yaks from Qinghai region were selected for the experiment.Blood samples were collected every 7 days from the date of delivery and divided into 5 groups based on time points:group A(delivery day),group B(7 days postpartum),group C(14 days postpartum),group D(21 days postpartum),and group E(28 days postpartum).Proteomic technology based on data independent scanning mode,protein mass spectra of each group was screened and compared pairwise.As a result,399 differentially expressed proteins were screened.And based on bioinformatics analysis,18 key proteins were identified,which were related to ovarian reproductive hormone secretion(PLA2,PKA,14-3-3,IGF1,IGFBP1,IGFBP4,IGFBP6,and IGFBP7),ovarian glucose metabolism and lipid metabolism(PFK,FBP,GAPDH,PK,LDHA,6PGDH,Apo IV,and LPL),as well as the other two key proteins(CBG and HSP9oβ).A network mechanism was constructed to regulate the reproductive system of yak postpartum key proteins.Through the analysis of key differentially expressed proteins,it is reflected that the abnormal secretion and metabolism of reproductive hormones in the ovaries of yaks after delivery,as well as the abnormal supply of energy from sugar metabolism and lipid metabolism, are the main reasons for the delayed recovery of the yak reproductive system after delivery.
