犊牛腹泻粪源弗格森埃希菌的分离鉴定及生物学特性研究

摘  要:为分离鉴定引起犊牛腹泻的致病菌,掌握其耐药性,从新疆额敏县某规模化牛场收集腹泻犊牛(19头)样本19份进行检测。采用细菌学培养和分子生物学技术分离鉴定弗格森埃希菌,对其进行超广谱β-内酰胺酶(ESBLs)表型、耐药性和毒力基因检测,并对其进行小鼠巨噬细胞的黏附与侵袭试验。结果显示,从19份粪便样本中分离到2株弗格森埃希菌(命名为292-1和337-1),均为产ESBLs弗格森埃希菌,基因型为blaTEM;药敏试验结果发现,2株分离菌对青霉素、头孢噻肟、利福平和氟苯尼考耐药,具有多重耐药性;耐药基因检测结果显示,2株弗格森埃希菌均携带blaTEM、floR、tetA、cmlA和sul2基因;毒力基因检测结果显示,2株弗格森埃希菌均携带yijP、iucD、ibeB、ompA和sodA毒力基因;细胞侵染试验结果显示,292-1和337-1对细胞的黏附率分别为3.23%和0.59%,侵袭率分别为0.12%和0.06%。In order to isolate and identify the causative organisms that cause diarrhoea in calves,and to grasp their drug resistance,19 samples of diarrhoeic calves(19)were collected from a large-scale cattle farm in Emin county,Xinjiang,and were tested.Escherichia fergusoni was isolated and identified by bacteriological culture and molecular biology techniques,and was subsequently tested for the presence of ultra-broad-spectrumβ-lactamases(ESBLs)phenotypes,drug resistance and virulence genes.It was also tested for adhesion and invasion by mouse macrophages.The results indicated that two strains of E.fergusonii were isolated from 19 faecal samples(designation 292-1 and 337-1),both of which were identified as ESBLs-producing E.fergusoni with the blaTEM genotype.Drug sensitivity tests conducted on 15 antibiotics revealed that the two isolates exhibited multi-drug resistant to penicillin,cefotaxime,rifampicin and florfenicol.The results of drug resistance gene testing showed that both strains of E.fergusoni carried the blaTEM,floR,tetA,cmlA and sul2 genes.Additionally,the results of virulence gene testing revealed that both strains possessed the yijP,iucD,ibeB,ompA and sodA virulence genes.The results of the cellular assay demonstrated an adhesion rate of 3.23%for 292-1 and 0.59%for 337-1,as well as an invasion rate of 0.12%and 0.06%,respectively.
