摘　　要：目的建立银黄肛肠洗液中盐酸小檗碱的含量测定方法。方法采用高效液相色谱法(HPLC法)测定。色谱柱(YMC-Triart C18,250 mm×4.6 mm,12 nm),柱温设定是25℃,将乙腈-0.1%磷酸溶液(50∶50)作为流动相,流速设置为1.0 ml/min;检测波长设置为265 nm;进样量选定为10μl。结果盐酸小檗碱在160.8~2010.0 ng范围内具有良好的线性关系,其线性方程Y=54274.15X-337814.38(n=5,r=0.9995);平均加样回收率为89.4%,相对标准偏差(RSD)为1.76%(n=6),精密度、重复性、稳定性符合定量检测要求。结论用HPLC法测定银黄肛肠洗液中盐酸小檗碱的含量,实际执行起来简捷、专属性较强、精密度较好、准确度高、稳定性好,值得推荐。Objective To establish a method for determining the content of berberine hydrochloride in Yinhuang anorectal lotion.Methods High-performance liquid chromatography(HPLC)was used for determination,with column(YMC-Triart C18,250 mm×4.6 mm,12 nm),column temperature of 25℃,acetonitrile-0.1%phosphoric acid solution(50∶50)as the mobile phase,flow rate of 1.0 ml/min,detection wavelength of 265 nm,sample size of 10μl.Results Berberine hydrochloride had a good linear relationship in the range of 160.8-2010 ng,and the linear equation was Y=54274.15X-337814.38(n=5,R=0.9995),the average recovery rate was 89.4%,the relative standard deviation(RSD)was 1.76%(n=6),and the precision,repeatability and stability met the requirements of quantitative detection.Conclusion The determination of berberine hydrochloride in Yinhuang anorectal lotion by HPLC has the advantages of simple operation,strong specificity,good precision,high accuracy and good stability,and is worthy of reference.
