摘 要:目的构建基于MOF-74@AuAg纳米花(NFs)的电化学生物传感器,并检测肺癌表皮生长因子受体(Egfr)基因突变的循环肿瘤DNA(ctDNA)。方法采用还原法制备AuAg NFs;采用溶剂热法制备金属有机框架材料MOF-74;利用壳聚糖的黏附性和成膜性制备AuAg NFs与MOF-74的复合材料;采用电子显微镜对AuAg NFs、MOF-74@AuAg NFs进行形貌表征;采用循环伏安法和电化学阻抗谱法评估构建的电化学生物传感器的可行性;采用差分脉冲伏安法定量检测Egfr基因突变的ctDNA,并采用加标回收法评估其在临床样本中的应用潜能。结果基于MOF-74@AuAg NFs的电化学生物传感器被成功构建,其对突变的ctDNA线性检测为10 fM~1 nM,检测限低至0.392 fM,加标回收率为92.22%~118.20%。结论本研究构建的电化学生物传感器可检测Egfr基因突变的ctDNA,其灵敏性高。Objective To construct an electrochemical biosensor based on MOF-74@AuAg nanoflowers(NFs),and detect circulating tumour DNA(ctDNA)with mutations in the epidermal growth factor receptor(Egfr)gene in lung cancer.Methods AuAg NFs were prepared by reduction method.Metal-organic framework material MOF-74 was prepared by solvothermal method.And composites of AuAg NFs and MOF-74 were prepared by utilizing the adhesive and film-forming properties of chitosan.AuAg NFs and MOF-74@AuAg NFs were characterized morphologically using electron microscope.Cyclic voltammetry and electrochemical impedance spectroscopy were used to evaluate the feasibility of the constructed electrochemical biosensor.Differential pulse voltammetry was used to quantitatively detect Egfr mutated ctDNA.And spiked recovery method was used to evaluate its potential application in real clinical samples.Results An electrochemical biosensor based on MOF-74@AuAg NFs was successfully constructed with a linear detection range of 10 fM to 1 nM for mutant ctDNA,a low detection limit of 0.392 fM,and a spiked recoveries range of 92.22%-118.20%.Conclusion The electrochemical biosensor constructed in this study can detect ctDNA of Egfr gene mutation with high sensitivity.
