摘  要:本文旨在实时检测针刺对离心运动性骨骼肌损伤后Ca^(2+)、Na^(+)、H_(2)O_(2)动态流速的影响,探讨针刺效应始动机制及信号转换途径。324只健康雄性Wistar大鼠随机分成6组:对照组、离心运动组(E)、E+针刺组(EA)、EA+瞬时受体电位(transient receptor potential,TRP)通道阻断剂组(EAT)、EA+NADPH氧化酶2 (nicotinamide adenine dinucleotide phosphate oxidase 2,NOX2)阻断剂组(EAN)和EA+PBS缓冲液安慰剂组(EAP)。经皮电刺激腓肠肌离心收缩后,即刻阻断剂预处理。30 min后,针刺内侧腓肠肌,采用非损伤微测技术实时检测留针期、拔针后即刻、3 h、6 h和24 h时相Ca^(2+)、Na^(+)、H_(2)O_(2)动态流速。结果显示:与E组相比,EA组Ca^(2+)净外流显著增加(P<0.05),同时Na^(+)净内流时相提前,伴随H_(2)O_(2)净外流大幅度下降(P<0.05)。然而,EAT和EAN组针刺效应显著下降,且H_(2)O_(2)过度净外流(P<0.001)。以上结果提示,针刺通过激活TRP通道协同NOX2活性,调控了离心运动性骨骼肌损伤后Na2+、Ca^(2+)和H_(2)O_(2)动态流速。This study aimed to investigate the effects of acupuncture on dynamic changes in Ca^(2+),Na^(+),and H_(2)O_(2) flux following eccentric exercise-induced muscle injury.The total of 324 healthy male Wistar rats were randomly divided into 6 groups:control group(C),eccentric exercise group(E),eccentric exercise with acupuncture group(EA),EA with TRP channel blocker group(EAT),EA with NOX2 blocker group(EAN)and EA with placebo group(EAP).Gastrocnemius muscles were subject to lengthening contractions with percutaneous electrical stimulation,followed by immediate pretreatment with blocking agents.After 30 min,acupuncture needling was administered to the gastrocnemius muscle,and real-time dynamic changes of Ca^(2+),Na^(+)and H_(2)O_(2) flux were measured with non-invasive micro-test technique during the needle retention period,immediately,3 h,6 h,and 24 h post-extraction respectively.Results showed that compared with the E group,acupuncture significantly increased net Ca^(2+)efflux(P<0.05),extended the period of net Na^(+)influx,and significantly decreased net H_(2)O_(2) efflux(P<0.05).However,these effects were significantly attenuated in the EAT and EAN groups,where excessive net H_(2)O_(2) efflux was observed(P<0.001).These findings indicate that acupuncture regulates the dynamic changes of Ca^(2+),Na^(+)and H_(2)O_(2) flux by activating the TRP channels and interacting with NOX2 activity following eccentric exercise-induced skeletal muscle injury.
