摘　　要：搭建了模块化、明-暗场模式可切换的显微成像系统,可用于透明生物细胞(如洋葱皮细胞)的暗场与明场无标记成像.基于该光子晶体器件的平面特性,将其与微流控芯片进行有机结合,实现了微流通道中物体运动行为的实时成像.通过搭建光学显微镜,并对采集的图像进行数据分析与计算,可以加深学生对菲涅耳公式、明-暗场成像原理、成像系统放大率、数值孔径等物理概念的理解,锻炼学生的实验动手能力和科研实践能力,开阔其眼界,提升其探索前沿科学的兴趣和能力.We had constructed a modular optical microscope whose bright and dark field modes could be switched conveniently.It could be used for imaging of transparent biological cells,such as the dark field and bright field imaging of the onion skin cells in a label-free manner.Owing to the planar nature of this photonic crystal device,we combined it with a microfluidic chip to achieve real-time imaging of the motion behavior of objects in microfluidic channels.By building an optical microscope with basic optical elements,together with the analysis and calculation for the data of the collected images,the students’understanding for physical concepts could be deepened,such as Fresnel’s formula,the bright-dark field imaging principle,the magnification of the imaging system,the numerical aperture,etc.And their hands-on experimental ability and scientific research practice ability would be exercised,their horizons would be broadened,and their interest in and ability to explore cutting-edge science could be enhanced as well.
