摘 要:采用单因素结合正交实验,对罗布麻叶总黄酮提取和纯化工艺分别进行优化,确定最佳条件。采用乙醇热回流提取法,提取罗布麻叶总黄酮,利用单因素实验和正交设计实验,对乙醇体积分数、提取温度、料液比、提取时间4个条件进行考察,在505 nm处测其吸光度,计算总黄酮提取率。选取三种(30-60、60-100、80-120目)不同型号聚酰胺树脂进行静态吸附解吸实验,选出最适合的树脂,通过单因素试验和正交试验分别对上样浓度、上样液pH、上样量、洗脱液体积进行考察,得到最佳分离纯化条件。结果表明,罗布麻叶总黄酮的最佳提取工艺条件是乙醇体积分数60%、提取温度是80℃、料液比1∶10、提取时间60 min。最佳分离工艺为上样浓度0.623 mg/mL、上样液pH=5、上样量为6 mL、洗脱液体积200 mL洗脱。乙醇回流提取法工艺简单、准确、重复性好,30-60目型聚酰胺吸附树脂对罗布麻叶总黄酮的静态吸附和解吸力最适合。The extraction and purification processes of total flavonoids from Apocynum venetum are optimized by single factor orthogonal experiment,and the optimal conditions are determined.The total flavonoids are extracted by ethanol reflux extraction method.Single factor experiment and orthogonal design experiment are used to investigate the ethanol volume fraction,extraction temperature,solid-liquid ratio and extraction time.The absorbance is measured at 505 nm,and the total flavonoids extraction rate is calculated.Three different types of polyamide resins(30-60,60-100,80-120 mesh)are selected for static adsorption and desorption experiments,and the most suitable resin is selected.The sample concentration,liquid pH,amount and elution liquid volume are investigated by single factor test and orthogonal test.The optimal separation and purification conditions are obtained.The results show that the optimal extraction conditions are 60%ethanol,80℃extraction temperature,1∶10 solid-liquid ratio and 60 min extraction time.The optimal separation process is as follows:0.623 mg/mL loading concentration,pH=5 loading liquid,6 mL loading volume,and 200 mL eluting liquid volume.The ethanol reflux extraction method is simple,accurate and reproducible,and the 30-60 mesh polyamide adsorption resin is the most suitable for static adsorption and suction of total flavonoids in the leaves.
