摘　　要：目的评估类风湿性关节炎(RA)患者血浆中中性粒细胞胞外诱捕网(NETs)的水平,以及NETs与RA指标之间的潜在关联。方法采用荧光染料法检测RA患者和健康对照组血浆DNA/NETs水平,ELISA法分析中性粒细胞弹性蛋白酶(NE)释放水平,并分析dsDNA和NE水平与ESR、CRP和ACPA的相关性;采用PMA体外刺激中性粒细胞生成NETs。结果与健康志愿者相比,RA患者血浆dsDNA/NETs水平显著升高,且活动期高于缓解期。PMA刺激后RA患者中性粒细胞生成NETs高于健康对照。dsDNA/NETs水平与ESR呈正相关;ACPA高水平(ACPA≥200U/mL)患者dsDNA/NETs及NE/NETs水平均明显高于低水平组(ACPA<200U/mL)。结论dsDNA/NETs与疾病活动性相关,dsDNA/NETs及NE/NETs与疾病病情相关。Objective To assess the levels of neutrophil extracellular trapping networks(NETs)in plasma of rheumatoid arthritis(RA)patients and the potential association between NETs and RA indicators.Methods Plasma DNA/NETs levels were measured in RA patients and healthy controls by fluorescent dye method,and neutrophil elastase(NE)release levels were analyzed by ELISA.dsDNA and NE levels were analyzed for their correlation with ESR,CRP,and ACPA;PMA was used to stimulate the production of NETs by neutrophils in vitro.Result:Compared with healthy volunteers,RA patients.Results Compared with healthy volunteers,plasma dsDNA/NETs levels were significantly higher in RA patients,and were higher in active than in remission.dsDNA/NETs levels in RA patients were higher than in healthy controls after PMA stimulation.dsDNA/NETs levels were positively correlated with ESR.dsDNA/NETs and NE/NETs levels in patients with high levels of ACPA(ACPA≥200 U/mL)were significantly higher than those in the group with low levels(ACPA<200U/mL).(ACPA<200 U/mL).Conclusion dsDNA/NETs were associated with disease activity,and dsDNA/NETs and NE/NETs were associated with disease condition.
