摘 要:目的 本研究旨在探讨多模态超声成像技术在微波消融(MWA)治疗裸鼠U87胶质瘤细胞移植瘤中的应用效果。方法 建立裸鼠U87胶质瘤细胞移植瘤模型,待瘤体长径达至1.2~1.5 cm时,在超声引导下对移植瘤进行MWA治疗。之后采用常规超声、弹性超声及超声造影(CEUS)等多模态成像技术,评估消融前及消融后肿瘤内部的各声学特征变化。随后,在消融后48 h对肿瘤进行病理检查,以评估消融效果并分析组织坏死情况。结果 本研究成功建立了12只裸鼠胶质瘤模型,MWA治疗后常规超声示瘤内回声显著增强,血流信号明显减少,CEUS示消融区几乎无造影剂充填,弹性成像表明瘤内硬度显著增加。消融后,瘤内二维超声灰度值从14.28±0.71显著增加至28.52±1.52(P<0.05),CEUS灰度值也从119.42±0.73降低至19.33±0.41(P<0.05)。病理检查结果表明,MWA治疗后消融区细胞坏死率达到94%。结论 多模态超声能够实时有效地评估裸鼠胶质瘤MWA治疗的效果,为MWA治疗脑肿瘤提供了初步的实验支持,并为进一步的临床应用提供了依据。Objective This study is aimed to investigate the application value of multimodal ultrasound imaging technology in the assessment of microwave ablation(MWA)treatment of U87 glioma cell transplanted tumors in nude mice.Methods A nude mouse U87 glioma cell transplanted tumor model was established.MWA treatment was per-formed on the transplanted tumors under ultrasound guidance when the tumor diameter reached 1.2-1.5 cm.Subse-quently,conventional ultrasound,elastography,and contrast-enhanced ultrasound(CEUS)were employed as multi-modal imaging techniques to assess the acoustic characteristics within the tumor before and after MWA.Pathological examination of the tumor was conducted 48 hours post-MWA to evaluate the MWA effect and to analyze the extent of tissue necrosis.Results The study successfully established 12 nude mouse glioma models.After MWA treatment,conventional ultrasound showed a significant increase in tumor echogenicity and a marked reduction in blood flow sig-nals.CEUS demonstrated that the MWA area was almost devoid of contrast medium filling,and elastography indica-ted a significant increase in tumor stiffness.Post-MWA,the gray-scale values within the tumor on two-dimensional ultrasound significantly increased from 14.28±0.71 to 28.52±1.52(P<0.05),and the CEUS gray-scale values also decreased from 119.42±0.73 to 19.33±0.41(P<0.05).Pathological examination results indicated that the cell necrosis rate in the ablation area reached 94%after MWA treatment.Conclusions Multimodal ultrasound imaging can effectively evaluate the effects of MWA treatment in nude mouse gliomas in real-time,providing preliminary experi-mental support for the use of MWA in the treatment of brain tumors and laying the groundwork for further clinical ap-plications.
