摘  要:本试验旨在探讨在暗斑发生率高的蛋鸡饲粮中添加有机锰、锌和纳米硒-褐藻寡糖对鸡蛋蛋壳暗斑发生的影响。试验选取288日龄罗曼粉蛋鸡150只(30只低暗斑发生率和120只高暗斑发生率),分为NC组(低暗斑+基础饲粮)、PC组(高暗斑+基础饲粮)、MnG组(高暗斑+基础饲粮+60 mg/kg羟基蛋氨酸锰+60 mg/kg MnSO_(4))、ZnG组(高暗斑+基础饲粮+55 mg/kg羟基蛋氨酸锌+55 mg/kg ZnSO_(4))和SeG组(高暗斑+基础饲粮+0.3 mg/kg纳米硒-褐藻寡糖),每组10个重复,每个重复3只鸡。试验期12周。结果表明:1)与NC组相比,PC组蛋壳强度、蛋壳水分含量和乳突层厚度显著增加(P<0.05),蛋壳钙含量、蛋壳膜粗蛋白质和总氨基酸含量显著降低(P<0.05),蛋壳腺钙结合蛋白-D28k(CaBP-D28k)mRNA相对表达量显著降低(P<0.05)。2)与PC组相比,各添加组鸡蛋暗斑评分平均值均显著降低(P<0.05)。3)与PC组相比,MnG组和SeG组蛋壳乳突层厚度均显著降低(P<0.05),蛋壳膜粗蛋白质含量显著增加(P<0.05)。此外,MnG和ZnG组蛋壳膜必需氨基酸和总氨基酸含量显著增加(P<0.05)。4)与PC组相比,MnG组蛋壳腺碳酸酐酶(CA)和CaBP-D28k mRNA相对表达量显著上调(P<0.05),ZnG组和SeG组蛋壳腺雌激素受体α(ERα)mRNA相对表达量显著上调(P<0.05)。综上所述,在暗斑发生率高的蛋鸡饲粮中添加120 mg/kg锰(60 mg/kg羟基蛋氨酸锰+60 mg/kg MnSO_(4))、110 mg/kg锌(55 mg/kg羟基蛋氨酸锌+55 mg/kg ZnSO_(4))或0.3 mg/kg纳米硒-褐藻寡糖,可能通过增加蛋壳钙含量和蛋壳膜粗蛋白质含量改善蛋壳结构,降低水分在蛋壳中的积聚,同时增加蛋壳腺钙代谢相关基因表达改善蛋壳腺功能,从而降低鸡蛋蛋壳暗斑的发生。The aim of this study was to explore the effects of dietary supplementation with organic manganese,zinc,nano-selenium alginate oligosaccharides on the occurrence of egg shell dark spots in laying hens with a high incidence of such spots.A total of 150 Roman pink laying hens at 288 days of age were selected,among which 30 had a low incidence of dark spots and 120 had a high incidence.They were divided into the NC group(low dark spot+basal diet),the PC group(high dark spot+basal diet),the MnG group(high dark spot+basal diet+60 mg/kg manganese hydroxymethionine+60 mg/kg MnSO_(4)),the ZnG group(high dark spot+basal diet+55 mg/kg zinc hydroxymethionine+55 mg/kg ZnSO_(4)),and the SeG group(high dark spot+basal diet+0.3 mg/kg nano-selenium alginate oligosaccharides),with 10 replicates per group and 3 chickens per replicate.The experiment lasted for 12 weeks.The results showed as follows:1)compared with the NC group,the eggshell strength,eggshell moisture content,and mastoid layer thickness in the PC group were significantly increased(P<0.05),while the eggshell calcium content,the crude protein content of the eggshell membrane,and the total amino acid content were significantly decreased(P<0.05).The relative mRNA expression of eggshell gland calcium-binding protein-D28K(CaBP-D28k)was significantly reduced(P<0.05).2)Compared with the PC group,the egg dark spot score mean in all the supplemented groups was significantly decreased(P<0.05).3)Compared with the PC group,the thickness of the eggshell mastoid layer in the MnG and SeG groups was significantly decreased(P<0.05),and the content of crude protein in the eggshell membrane was significantly increased(P<0.05).Additionally,the contents of essential amino acids and total amino acids of the eggshell membrane in the MnG and ZnG groups were significantly increased(P<0.05).4)Compared with the PC group,the relative mRNA expression levels of carbonic anhydrase(CA)and CaBP-D28k in the eggshell gland in the MnG group were significantly up-regulated(P<0.05),and the relative mRNA e
