摘 要:本研究旨在通过构建氧化应激动物模式,评估白头翁口服液(BTW)对肉鸡免疫和抗氧化能力的影响。本试验选用21日龄无特定病原体(SPF)雄性白来航鸡30只,随机分到3个组,分别为对照组(CON组)、脂多糖(LPS)组(LPS组)和BTW+LPS组(B-L组),每组10个重复,每个重复1只鸡。CON组在31、33和35日龄腹腔注射2 mL生理盐水;LPS组在31、33和35日龄腹腔注射LPS(0.5 mg/kg BW);B-L组在21~35日龄每天对每只鸡灌服4 mL BTW,并在31、33和35日龄腹腔注射LPS(0.5 mg/kg BW),试验期为14 d。结果表明:与CON组相比,LPS组鸡出现明显腹泻症状,血清活性氧(ROS)和丙二醛(MDA)的含量显著升高(P<0.05),血清总抗氧化能力(T-AOC)显著降低(P<0.05),表明LPS诱导的SPF鸡氧化应激模型建立成功。免疫指标结果显示,与LPS组相比,摄入BTW显著升高了模型鸡血液血红蛋白(HGB)水平和红细胞数量(RBC)(P<0.05),显著降低了模型鸡血清肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)的含量(P<0.05),显著升高了血清白细胞介素-4(IL-4)和白细胞介素-10(IL-10)的含量(P<0.05),并维持在与CON组近似水平(P>0.05)。抗氧化指标结果显示,与LPS组相比,摄入BTW显著升高了模型鸡血清抗氧化酶中谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)、超氧化物歧化酶(SOD)活性(P<0.05);与此同时,BTW还维持了模型鸡回肠黏膜形态、肝脏和免疫器官指数的正常。综上所述,本研究证实BTW可以通过调节家禽体内炎症细胞因子含量和抗氧化酶活性,以及影响组织器官的发育,从而改善了模型鸡免疫和抗氧化能力,进而缓解了模型鸡肠道炎症和氧化应激反应。The aim of this study was to construct oxidative stress model animals by attacking specific pathogen free(SPF)white Leghorn chickens,and then evaluate the effects of Baitouweng oral liquid(BTW)on the immune and antioxidant capacity of broilers in poultry production.Thirty 21-day-old SPF male white Leghorn chickens were randomly assigned to 3 groups:control group(CON group),lipopolysaccharide(LPS)group(LPS group)and BTW+LPS group(B-L group)with 10 replicates per group and 1 chicken per replicate.In CON group,2 mL normal saline was injected intraperitoneally at 31,33 and 35 days of age.The LPS groups were intraperitoneally injected with LPS(0.5 mg/kg of BW)at 31,33 and 35 days of age.Chickens in the B-L group were given 4 mL BTW per day at 21 to 35 days of age,and LPS(0.5 mg/kg of BW)was intraperitoneally injected at 31,33 and 35 days of age.The experiment lasted for 14 days.The results showed that compared with CON group,LPS group showed obvious diarrhea symptoms,serum reactive oxygen species(ROS)and malondialdehyde(MDA)contents were significantly increased(P<0.05),and total antioxidant capacity(T-AOC)was significantly decreased(P<0.05),indicating that the oxidative stress model of SPF chickens induced by LPS was successfully established.Immune index results showed that compared with LPS group,BTW intake significantly increased blood hemoglobin(HGB)level and red blood cell count(RBC)of model chickens(P<0.05).The contents of tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β)and interleukin-6(IL-6)in serum of model chickens were significantly decreased(P<0.05).The contents of interleukin-4(IL-4)and interleukin-10(IL-10)in serum were significantly increased(P<0.05),and were maintained at similar levels to those in CON group(P>0.05).The results of antioxidant indexes showed that compared with LPS group,the activities of glutathione peroxidase(GSH-Px),catalase(CAT)and superoxide dismutase(SOD)in serum of model chickens were significantly increased by BTW intake(P<0.05).At the same time,BTW also maintained the nor
