摘 要:本试验旨在研究饲粮中添加羧甲基茯苓多糖(CMP)、丁酸梭菌和罗伊氏乳杆菌后生元(PB)复合物对昆明小鼠生长性能和盲肠菌群结构的影响。选取3周龄、体重10 g左右昆明雄性小鼠24只,随机分为4组(每组6个重复,每个重复1只):对照组饲喂基础饲粮,试验组分别饲喂在基础饲粮中添加1‰CMP(1‰CMP组)、5‰PB(5‰PB组)和6‰复合物(1‰CMP+5‰PB,6‰复合物组)的饲粮。试验期13 d。结果表明:1)2和10 mg/mL的CMP对羟自由基和1,1-二苯基-2-三硝基苯肼(DPPH)自由基的清除率分别为49.83%和52.89%。然而即使10 mg/mL的CMP也只有微弱的还原力,对超氧自由基没有任何清除效果。2)采用两步发酵法制备PB,第一步发酵液中丁酸梭菌和第二步发酵液中罗伊氏乳杆菌含菌量均为5×10^(8) CFU/mL。CMP与PB复合物配伍后,其中CMP、PB、麦芽糊精和轻质碳酸钙辅料的质量百分比为25∶5∶19∶1。3)饲喂13 d后,与1‰CMP组和5‰PB组对比,6‰复合物组小鼠的平均日采食量分别增加8.65%和11.44%,平均日增重分别增加12.42%和8.18%,体重分别增加了7.37%和4.63%。4)肠道菌群测序结果表明,6‰复合物促进昆明小鼠体重增长的机理可能与厚壁菌门(Firmicutes)/拟杆菌门(Bacteroidetes)(F/B)的比值显著升高(P<0.05)有关,并且小鼠盲肠内容物中螺杆菌属(Helicobacter)和肠鼠杆菌(Muribaculum intestinale)等有害菌相对丰度也显著降低(P<0.05),而亨盖特丁酸弧菌(Butyrivibrio hungatei)等有益菌相对丰度显著升高(P<0.05)。线性判别分析效应量(LEfSe)分析也提示6‰复合物组小鼠肠道中有益菌群显著富集。综上所述,CMP有较强的清除羟自由基的能力,添加6‰复合物(1‰CMP+5‰PB)可能通过提高肠道中F/B的比值和改善肠道菌群结构提升昆明小鼠的生长性能。The objective of this research was to examine the impacts of a compound comprising carboxymethylpachymaran(CMP)and postbiotics(PB)derived from Clostridium butyricum and Lactobacillus reuteri on the growth performance and cecal microflora of Kunming mice.Twenty-four male Kunming mice,aged 3 weeks and weighing approximately 10 g,were randomly assigned to four groups with 6 replicates per group and 1 mouse per replicate.The control group received a basal diet,while the experimental groups were fed the basal diet supplemented with 1‰CMP(1‰CMP group),5‰PB(5‰PB group)and 6‰ compound(1‰CMP+5‰PB,6‰ compound group),respectively.The experiment lasted for 13 days.The results showed as follows:1)CMP at concentrations of 2 and 10 mg/mL exhibited scavenging rates of 49.83% and 52.89% for hydroxyl and 1,1-diphenyl-2-trinitrophenylhydrazine(DPPH)free radicals,respectively.Despite this,even at a concentration of 10 mg/mL,CMP demonstrated only limited reducing power and no scavenging activity against superoxide free radicals.2)The postbiotics were produced using a two-step fermentation process,resulting in a concentration of 5×108 CFU/mL of viable Clostridium butyricum in the initial step and Lactobacillus reuteri in the subsequent step.The composition of CMP,PB,maltodextrin,and light calcium carbonate excipients in the complex was determined to be 25∶5∶19∶1 following the mixing of CMP with PB.3)After a 13-day feeding period,it was observed that the average daily feed intake of mice increased by 8.65% and 11.44% when compared with the 1‰CMP group and 5‰PB group,respectively.Additionally,the average daily gain showed an increase of 12.42% and 8.18% in the same groups,while body weight exhibited a 7.37% and 4.63% increase,respectively.4)The findings from the intestinal microflora sequencing analysis indicate that the compound’s mechanism for promoting weight gain in Kunming mice may be associated with a notable elevation in the Firmicutes to Bacteroides(F/B)ratio(P<0.05).Additionally,there was a signific
