摘 要:目的:探讨地稔抗Hp相关慢性非萎缩性胃炎作用。方法:60只SD大鼠分为正常对照组、模型组、阳性药物组和地稔低、中、高剂量组,每组10只。除正常对照组外,其余采用阿莫西林+Hp灌胃方法建模。正常对照组和模型组大鼠每天灌胃生理盐水,阳性组灌胃阿莫西林(100 mg/kg),地稔组灌胃地稔水煎液400、2 000、4 000 mg/kg,连续灌胃14 d。ELISA法检测大鼠血清IL-1β、IL-2、TNF-α、IFN-β、CRP,透射电镜检测大鼠胃黏膜组织病理变化,qRT-PCR检测大鼠胃黏膜组织中炎症因子和Hp基因表达,Western Blotting检测大鼠胃黏膜组织炎症因子蛋白。结果:地稔中、高剂量组可以显著降低大鼠血清IL-1β、IL-2、TNF-α、IFN-β、CRP含量,地稔高剂量组可以显著降低大鼠胃黏膜组织IL-1β、IL-2、TNF-α、IFN-β、CRP和尿素酶mRNA表达水平和蛋白表达量,地稔低、中、高剂量组可以改善大鼠胃黏膜组织病理变化。结论:地稔具有抗大鼠Hp相关性慢性非萎缩性胃炎作用,其作用机制可能与抑制炎症因子相关。Objective:To investigate the antagonism effect of Melastoma dodecandrum on Hp-related chronic non-atrophic gastritis.Methods:60 SD rats were divided into normal control group,model group,positive group,low,medium and high dose of Melastoma dodecandrum groups with 10 rats in each group.Except thecontrol group,the rest rats were modeled by amoxicillin+Hp gavage.Rats in normal group and model group were intragastrically given saline every day,the positive group was intragastrically given Amoxicillin at 100 mg/kg,and the Melastoma dodecandrum groups were intragastrically given 400,2000,4000 mg/kg Melastoma dodecandrum water decoction,respectively,for 14 days.The contents of IL-1β,IL-2,TNF-α,IFN-β,CRP in serum of rats were detected by ELISA,the pathological changes of gastric mucosa were detected by transmission electron microscopy,and the expression levels of inflammatory factors and Hp gene in gastric mucosa were detected by real-time fluorescence quantitative PCR.Western Blotting was used to detect inflammatory factor proteins in gastric mucosa of rats.Results:The contents of IL-1β,IL-2,TNF-α,IFN-β,CRP in serum of rats could be significantly decreased in medium and high dose of Melastoma dodecandrum groups,while the mRNA and protein expression levels of IL-1β,IL-2,TNF-α,IFN-β,CRP and urease in gastric mucosa of rats were significantly decreased in high dose of Melastoma dodecandrum group.The pathological changes of gastric mucosa in low,medium and high doses of Melastoma dodecandrum were improved.Conclusion:Melastoma dodecandrum can effectively treat HP-related chronic non-atrophic gastritis in rats,and its mechanism may be related to inhibiting inflammatory factors.
