重楼皂苷Ⅰ对肺癌A549细胞增殖、细胞周期、迁移、侵袭及代谢物的影响

摘　　要：目的:研究重楼皂苷Ⅰ对A549细胞增殖、细胞周期、迁移和侵袭的抑制作用,并基于迁移相关蛋白和单细胞代谢检测探讨其作用机制。方法:采用不同浓度重楼皂苷Ⅰ处理人肺癌A549细胞,使用CCK-8检测细胞毒性及增殖抑制作用;流式细胞术检测细胞周期;细胞划痕实验和Tranwell侵袭实验检测细胞迁移抑制作用;Western blot检测细胞迁移和细胞粘连相关蛋白(P-p38、Slug、β-catenin、Claudin-1、N-Cadherin、MMP-2)表达;单细胞质谱检测细胞代谢物差异。结果:重楼皂苷Ⅰ可明显抑制A549细胞的增殖和迁移,显著降低P-p38,Slug,β-catenin,Claudin-1和N-Cadherin、MMP-2蛋白表达以及脂类代谢物水平。结论:重楼皂苷Ⅰ可以抑制A549细胞增殖、迁移、侵袭,降低细胞迁移和细胞粘连相关蛋白表达和细胞代谢水平。Objective:To study the inhibitory effect of Polyphyllin I on the proliferation,cell cycle,migration and invasion of non-small cell lung cancer A549 cells,and to explore its mechanism based on the detection of migration-related proteins and single-cell metabolism.Methods:Human lung cancer A549 cells were cultured with different concentrations of Polyphyllin I,and the cytotoxicity and proliferation inhibition were detected by CCK-8.Cell cycle was detected by flow cytometry.Cell migration inhibition was detected by cell scratch assay and Tranwell invasion assay.Western blot was used to detect the expressions of cell migration and cell adhesion related proteins(P-p38,Slug,β-catenin Claudin-1,N-Cadherin,MMP-2).The difference of cell metabolites was detected by single cell mass spectrometry.Results:Polyphyllin I could significantly inhibit the proliferation and migration of non-small lung cancer A549 cells,and significantly reduce the protein expressions of P-p38,Slug,β-catenin,Claudin-1,N-Cadherin and MMP-2,as well as the lipid metabolites.Conclusion:Polyphyllin I can inhibit A549 cell proliferation,migration,invasion,reduce the expressions of cell migration and cell adhesion related proteins and cell metabolism.
