摘 要:目的:探讨熊果酸(UA)通过调节circ_0090231与circ_0004543表达对氧化低密度脂蛋白(ox-LDL)诱导血管平滑肌细胞(VSMCs)损伤的保护作用。方法:实验分4组:空白对照组(Control),氧化低密度脂蛋白组(ox-LDL),熊果酸预处理组(UA)和ox-LDL+熊果酸处理组(UA+ox-LDL),CCK8测定细胞活力值、EDU测定细胞增殖、划痕实验测定细胞迁移行为,RT-PCR法测定circ_0090231与circ_0004543表达。结果:UA预处理可显著减轻ox-LDL诱导的VSMC细胞损伤。在UA预处理组中,细胞活力值、细胞增殖率显著降低(均P<0.05),迁移能力下降(P<0.05)。RT-PCR法显示:ox-LDL组相较于Control组,circ_0090231与circ_0004543表达均上调(均P<0.05),UA+ox-LDL组相较于ox-LDL组,circ_0090231与circ_0004543表达均下调(均P<0.05)。结论:熊果酸通过下调circ_0090231与circ_0004543表达保护VSMCs免受ox-LDL诱导的损伤,显示出其在心血管疾病治疗和预防中有潜在的应用价值。Objective:To explore the potential therapeutic effects of Ursolic Acid(UA)on protecting vascular smooth muscle cells(VSMCs)from damage induced by oxidized low-density lipoprotein(ox-LDL)by regulating the expressions of circ_0090231 and circ_0004543.Methods:VSMCs were divided into 4 groups:Control group,Oxidized Low-Density Lipoprotein group(ox-LDL),Ursolic Acid pretreatment group(UA),and ox-LDL+Ursolic Acid treatment group(UA+ox-LDL).Cell viability was assessed using the CCK-8 assay,cell proliferation was measured by EDU assay,and cell migration was evaluated through scratch tests.The expression levels of circ_0090231 and circ_0004543 were determined by RT-PCR.Results:UA pretreatment significantly alleviated ox-LDL-induced damage in VSMCs.In the UA pretreatment group,cell viability and proliferation rates significantly decreased(both P<0.05),and migration capability was reduced(P<0.05).RT-PCR results showed that compared to the control group,the expressions of circ_0090231 and circ_0004543 were upregulated in the ox-LDL group(both P<0.05),while in the UA+ox-LDL group compared to the ox-LDL group alone,the expressions of both circular RNAs was downregulated(both P<0.05).Conclusion:Ursolic Acid protects VSMCs from ox-LDL-induced damage by downregulating the expressions of circ_0090231 and circ_0004543,highlighting its potential application in the treatment and prevention of cardiovascular diseases.
