冬眠期和活跃期密点麻蜥不同器官的抗氧化应激及肠道菌群比较

摘  要:氧化应激是调节种群适应的重要生态和进化驱动力,并可能与动物的肠道菌群密切相关。为探究冬眠期蜥蜴不同器官氧化应激的特异性及肠道菌群的可能作用,比较了冬季(冬眠期)和夏季(活跃期)密点麻蜥Eremias multiocellata的脑、肾脏、小肠和前肢肌肉中抗氧化酶的活性及肠道菌群的组成及多样性。结果显示,夏季密点麻蜥的脑、肾脏和骨骼肌的丙二醛含量显著高于冬季,且脑和骨骼肌的超氧化物歧化酶、肾脏和骨骼肌的谷胱甘肽过氧化酶活性高于冬季,但脑和肾脏的过氧化氢酶活性低于冬季。夏季肠道菌群的α多样性和拟杆菌门Bacteroidetes的相对丰度低于冬季,但放线菌门Actinobacteria的相对丰度高于冬季;夏季与抗氧化应激有关的通路,如丁酸酯代谢、脂肪酸降解和硫代谢的相对丰度上调。这表明密点麻蜥在季节适应中各种抗氧化酶在不同器官的表现并不完全相同,且肠道菌群可能参与了小肠的抗氧化。该结果有助于认识荒漠区冬眠蜥蜴不同器官的抗氧化应激。Oxidative stress is an important ecological and evolutionary driver regulating population adaptation and may be closely related to gut microbiota.To explore the specificity of oxidant stress in various organs of lizards during the hibernation period and the possible role of gut microbiota,the antioxidant enzyme activities in the brain,kidneys,small intestine,and forelimb muscles and gut microbiota diversity in the Eremias multiocellata between winter(hibernation)and summer(active period)were compared.The results showed that the contents of malondialdehyde in the brain,kidneys,and skeletal muscles of E.multiocellata in summer were significantly higher than those in winter,and the activities of superoxide dismutase in the brain and skeletal muscles,glutathione peroxidase in the kidneys and skeletal muscles were significantly higher than those in winter.The catalase activities of the brain and kidneys in summer were significantly lower than those in winter.The alpha diversity and relative abundance of Bacteroidetes in the gut microbiota in summer were significantly lower than those in winter,whereas the relative abundance of Actinobacteria was higher than that in winter.The pathways associated with resistance to oxidative stress,e.g.relative abundance of butanoate metabolism,fatty acid degradation and sulphur metabolism were up-regulated in summer.These results suggested that the performance of various antioxidant enzymes in different organs of lizards is not entirely the same during seasonal adaptation,and the gut microbiota may be involved in the antioxidant defense of the small intestine.This study contributes to understanding the antioxidant stress of different organs of hibernating lizard in desert.
