摘 要:白星花金龟Protaetia brevitarsis幼虫是重要的农林害虫和经济昆虫。采用保幼激素(JH)和蜕皮激素(20E)处理白星花金龟幼虫,并通过GeNorm、NormFinder、ΔCt法和BestKeeper等方法评估了8个候选内参基因(Rps3、EF-1α、27A、AK、Act、GAPDH、Rps18、Tub)的表达稳定性。根据RefFinder的综合排名,Rps3、Rps18和EF-1α分别在JH、发育阶段和20E处理下表现出较高的稳定性,适合作为白星花金龟的内参基因;JH和20E水平的变化会影响内参基因的表达稳定性。研究结果为白星花金龟基因表达研究提供了可靠的内参基因参考,为农业生产及废物资源化利用提供了理论依据;另一方面,筛选的内参基因可支持JH类似物相关实验的数据标准化,为成虫防控提供帮助。The larva of white spotted flower chafer(Protaetia brevitarsis)holds significant importance both as an agricultural and forestry pest and as an insect with high economic value.In this study,the larvae were subjected to treatments with juvenile hormone(JH)and 20-hydroxyecdysone(20E),and the expression stability of 8 candidate reference genes(Rps3,EF-1α,27A,AK,Act,GAPDH,Rps18,and Tub)was evaluated using GeNorm,NormFinder,ΔCt,and BestKeeper.The results showed that,according to the comprehensive ranking index provided by RefFinder,Rps3,Rps18,and EF-1αdemonstrated remarkable stability under the treatment of JH,developmental stage and 20E,respectively,thereby qualifying them as reference genes for this species.Furthermore,this study also uncovered that alterations in the concentrations of JH and 20E could exert an influence on the expression stability of these reference genes.Consequently,the selection of appropriate reference genes under conditions involving hormone treatments is crucial for improving the accuracy of gene expression analysis.This study provides a set of reliable reference genes for conducting gene expression studies in P.brevitarsis and may lay a foundation for agricultural production and waste resource utilization.Additionally,the identified reference genes serve as valuable tools for data normalization in experiments involving JH analogs,thereby facilitating the control of adult pests.
