摘 要:试验旨在研究黄连提取物(CCE)对葡聚糖硫酸钠(DSS)诱导的小鼠溃疡性结肠炎(UC)的防治作用。选择60只C57BL/6小鼠,随机分成6组,即对照组(CON)组、DSS模型组、美沙拉嗪(MES)阳性对照组(200 mg/kg)及CEE低(CEE-L,600 mg/kg)、中(CEE-M,1000 mg/kg)、高(CEE-H,1400 mg/kg)剂量组。预试期7 d,正式试验期14 d。结果显示,与DSS组相比,CEE组极显著抑制了UC小鼠的体重降低(P<0.01),DAI评分和结肠长度极显著增加(P<0.01)。CEE组改善了UC小鼠结肠组织病理损伤的情况,炎性细胞浸润减少,肠绒毛和隐窝结构有所恢复。与DSS组相比,CEE组结肠中白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)mRNA的表达水平极显著下调(P<0.01)。与DSS组相比,在门水平,CEE组变形菌门(Proteobacteria)、弯曲菌门(Campylobacter)相对丰度明显降低,CEE-L组、CEE-M组疣微菌门(Verrucomicrobacteria)的相对丰度明显增加,CEE-M组、CEE-H组拟杆菌门(Bacteroidetes)、脱硫杆菌门(Desulphurobacteria)的相对丰度明显增加;在属水平,CEE-M组、CEE-H组大肠杆菌志贺菌属(Escherichia-Shigella)的相对丰度明显降低,CEE-L组、CEE-M组阿克曼氏菌属(Ackermania)的相对丰度明显升高。研究表明,CCE可通过抑制炎症因子表达和维持肠道菌群稳态发挥对UC小鼠的预防保护作用。The study aimed to explore the prevention and treatment effect of Coptischinensisextract(CCE)on ulcerative colitis(UC)induced by sodium dextranate sulfate(DSS)in mice.Six groups of sixty C57BL/6 mice were randomly selected,namely control group(CON),DSS model group,mesalazine(MES)positive control group(200 mg/kg),low CEE(CEE-L,600 mg/kg),medium CEE(CEE-M,1000 mg/kg),high CEE(CEE-H,1400 mg/kg)dose group.The pre-test period was seven days and the formal test preiod was 14 days.The results showed that compared with DSS group,CCE group extremely inhibited weight loss(P<0.01),DAI score and colon length of UC mice were extremely increased(P<0.01).CEE group improved the pathological injury of colon tissue in UC mice,the intestinal villi and crypt structure were restored,and there was a decrease in the invasion of inflammatory cells.Compared with DSS group,the expression levels of interleukin-1β(IL-1β),interleukin-6(IL-6),and tumor necrosis factor-α(TNF-α)mRNA in colon of CEE group were extremely down-regulated(P<0.01).At the phylum level,there was a substantial decrease in the relative abundance of Proteobacteria and Campylobacter in the CEE group when compared to the DSS group.There was a notable rise in the relative abundance of Verrucomicrobacteria in the CEE-L and CEE-M groups,and the relative abundance of Bacteroidetes and Desulphurobacteria in CEE-M and H groups were significantly increased.In terms of genus level,in the CEE-M and CEE-H groups,there was a notable decrease in the relative abundance of Escherichia-Shigella,but an apparent rise in the relative abundance of Ackermaniain the CEE-L and CEE-M groups.The study indicates that CCE can play a preventive and protective role in UC mice by inhibiting the expression of inflammatory factors and maintaining the homeostasis of intestinal flora.
