摘 要:试验旨在研究柱花草自然青贮发酵品质及其青贮饲料中梭菌的分离与鉴定。选取青贮60 d后的柱花草作为研究对象,测定其发酵品质及营养成分,并利用传统培养法和16S rDNA序列分析对青贮饲料中分离的梭菌进行鉴定。结果显示,营养生长期柱花草自然青pH值为5.31、氨态氮/总氮为39.39%,丁酸含量为0.77%;从柱花青贮饲料中分离得到1株梭菌,经形态学观察、生理生化测试和分子生物学鉴定为丁酸梭菌并命名为4233,该梭菌在15℃和pH值3.5时分别有最小光密度值(OD_(600 nm)),为0.026和0.223,在35~45℃时OD_(600 nm)值均高于1.600,在pH值7.5~9.0时OD_(600 nm)值在1.600左右。研究表明,柱花草自然青贮是典型的梭菌发酵,青贮饲料中的梭菌是丁酸梭菌。The study aimed to investigate the fermentation quality of natural stylo silage and isolation and identification of Clostridiumfrom silage.Ensiling stylo 60 days was selected as research material,and its fermentation quality and nutrient composition were determined,and using conventional cultivation methods and 16S rDNA sequence analysis to identify isolated Clostridium in silage.The results showed that the vegetative stage of natural stylo silage pH value was 5.31,ammonia nitrogen/total nitrogen was 39.39%and butyric acid content was 0.77%of dry matter.A strain of Clostridium butyricum was isolated from stylo silage,which was identified by morphological observation,physiological and biochemical tests,molecular biological identification and named Clostridium butyicum 4233.The minimum optical density values(OD_(600nm))of strain Clostridiumbutyricum 4233 were 0.026 and 0.223 at 15℃and pH 3.5,respectively.The OD_(600nm) values were higher than 1.600 at 35~45℃,and the OD_(600nm) values were about 1.600 at pH value 7.5~9.0.The study indicates that the natural silage of stylo was a typical clostridial fermentation,and clostridial species in the silage was Clostridiumbutyricum.
