摘 要:本研究建立了一种对半胱氨酸、壳寡糖和不同分子量壳聚糖精确定量的紫外分光光度法。通过对定量波长、溶剂类型、共存离子、pH、溶液保存温度和时间等因素的对比分析,确定了半胱氨酸、壳寡糖和壳聚糖定量的最佳条件。结果表明,以水溶解半胱氨酸和壳寡糖,以高氯酸溶解壳聚糖,在室温(25℃),pH在2~5的范围内,半胱氨酸、壳寡糖以及不同分子量壳聚糖分别在191~197 nm波长处浓度与吸光度相关性最佳(标准曲线的相关系数达0.999以上,且线性范围较宽);该方法不易受pH、保存时间和温度等影响,且结果准确稳定,回收率近100%。在操作便捷性、结果准确性和稳定性上均有较好表现。In this study,a precise quantification method using a UV spectrophotometric technique for cysteine,chitooligosaccharides,and chitosan with different molecular weights was developed.The optimal conditions for quantification were determined by considering factors such as wavelength,solvents,presence of coexisting anions and cations,pH,solution storage temperature and time.In a pH range of 2~5,the best correlation of concentration-absorbance was found for cysteine,chitooligosaccharides,and various molecular weight chitosan(solved in perchloric acid)at 191~197 nm,respectively(the correlation coefficients of the standard curve equations were greater than 0.999,and the linear ranges were wide);With the UV spectrophotometric method,the accuracy and stability in determining results with recovery rates close to 100%.It also exhibited less sensitivity to pH or temperature.
