摘 要:本研究采用热水浸提法提取黑木耳多糖并对其单糖组成进行分析。通过单因素考察和正交实验优化提取工艺,确定最佳工艺参数为提取时间3h、液料比为40∶1(mL∶g)、提取温度100℃及提取次数3次,在此工艺条件下,黑木耳多糖的平均得率为16.69%。采用高效液相色谱法指认黑木耳降解多糖中的单糖成分并对其进行定量分析,结果显示,黑木耳多糖降解后的主要单糖为乙酰葡萄糖、蔗糖、葡萄糖和木糖,含量分别为48.12%、18.13%、10.19%和3.21%。本研究可为黑木耳多糖的进一步开发和利用提供科学参考。This study used hot water extraction method to extract polysaccharides from black fungus and analyzed their monosaccharide composition.Through single factor investigation and orthogonal experiment optimization of the extraction process,the optimal process parameters were determined as extraction time of 3 hours,liquid to material ratio of 40∶1(mL∶g),extraction temperature of 100℃and extraction times of 3 times.Under these process conditions,the average yield of black fungus polysaccharides was 16.69%.Using high-performance liquid chromatography,the monosaccharide components in the degraded polysaccharides of black fungus were identified and quantitatively analyzed.The results showed that the main monosaccharides after the degradation of black fungus polysaccharides were acetyl glucose,sucrose,glucose and xylose,with contents of 48.12%,18.13%,10.19%and 3.21%,respectively.This study can provide scientific reference for the further development and utilization of polysaccharides from black fungus.
